Abstract

Disclosed herein are compositions comprising a serotonergic tryptamine compound, combinations thereof, and methods of using them for treating and preventing a variety of human conditions. In one embodiment, disclosed herein are new compositions and methods which comprise a therapeutically effective amount of a first purified psilocybin derivative and a therapeutically effective amount of a second compound. The second compound is selected from the group consisting of a second purified psilocybin derivative, a purified terpene, a serotonergic drug, an adrenergic drug, a dopaminergic drug, a purified erinacine, and a purified hericenone.

Claims (23)

1. A dosage composition comprising a homogenous mixture of:

a first psilocybin derivative selected from [3-(2-trimethylaminoethyl)-1H-indol-4-yl] dihydrogen phosphate, 4-hydroxy-N,N,N-trimethyltryptamine, [3-(2-methylaminoethyl)-1H-indol-4-yl] dihydrogen phosphate, 4-hydroxy-N-methyltryptamine, [3-(aminoethyl)-1H-indol-4-yl] dihydrogen phosphate, 4-hydroxytryptamine, and their salts; and

a second compound, wherein the second compound is an 80-100% pure erinacine selected from the group consisting of erinacine A, erinacine B, erinacine C, erinacine D, erinacine E, erinacine F, erinacine G, erinacine H, erinacine I, erinacine J, erinacine K, erinacine L, erinacine P erinacine Q, and erinacine R or an 80-100% pure hericenone selected from the group consisting of hericenone A, hericenone B, hericenone C, hericenone D, hericenone E, hericenone F, hericenone G, and hericenone H;

wherein the dosage composition comprises a molar ratio of between about 100:1 to about 1:100 of the first psilocybin derivative and the second compound;

wherein the pure erinacine or the pure hericenone represents 80-100% by mass of the total erinacine or hericenone in the dosage composition; and

wherein the dosage composition further comprises a monoamine oxidase inhibitor.

2. The dosage composition of claim 1 , further comprising an excipient.

3. The dosage composition of claim 1 , wherein the composition comprises a molar ratio of between about 10:1 to about 1:10 of the first psilocybin derivative and the second compound.

4. The dosage composition of claim 1 , wherein the composition comprises a molar ratio of between about 5:1 to about 1:5 of the first psilocybin derivative and the second compound.

5. The dosage composition of claim 1 , wherein the composition comprises a molar ratio of between about 50:1 to about 1:50 of the first psilocybin derivative and the second compound.

6. The dosage composition of claim 1 , wherein the composition comprises a molar ratio of between about 75:1 to about 1:75 of the first psilocybin derivative and the second compound.

7. The dosage composition of claim 1 , wherein the first psilocybin derivative comprises [3-(2-methylaminoethyl)-1H-indol-4-yl] dihydrogen phosphate.

8. The dosage composition of claim 1 , wherein the first psilocybin derivative comprises 4-hydroxy-N-methyltryptamine.

9. The dosage composition of claim 8 , wherein the 4-hydroxy-N-methyltryptamine is a freebase.

10. The dosage composition of claim 9 , wherein the 4-hydroxy-N-methyltryptamine freebase is crystalline.

11. The dosage composition of claim 8 , wherein the 4-hydroxy-N-methyltryptamine is a crystalline salt.

12. The dosage composition of claim 1 , wherein the second compound is a 90-100% pure erinacine or a 90-100% pure hericenone, wherein the pure erinacine or the pure hericenone represents 90-100% by mass of the total erinacine or hericenone in the dosage composition.

13. The dosage composition of claim 1 , wherein the chemical composition is determined by chromatography and/or spectrometry.

14. The dosage composition of claim 1 , further comprising pyridine-3-carboxylic acid.

15. The dosage composition of claim 1 , wherein the dosage composition comprises 0-5 mass percent of water.

16. The dosage composition of claim 1 , wherein the second compound is erinacine C.

17. The dosage composition of claim 1 , wherein the dosage composition is substantially free from 4-hydroxy-N,N-dimethyltryptamine.

18. The dosage composition of claim 1 , wherein the dosage composition is substantially free from [3-(2-Dimethylaminoethyl)-1H-indol-4-yl] dihydrogen phosphate.
