Fc variants with altered binding to FcRn
The present application relates to a variant Fc region comprising at least one modification relative to a wild-type human Fc region, where the modification selected from the group consisting of 434S, 252Y/428L, 252Y/434S, and 428L/434S, and the numbering is according to the EU index.
1. An anti-TNF antibody comprising:
a. two identical heavy chain polypeptides, each having the amino acid sequence of SEQ ID NO: 29; and
b. two identical light chain polypeptides, each having the amino acid sequence of SEQ ID NO: 23.
2. A nucleic acid comprising:
a. a first polynucleotide encoding a heavy chain polypeptide having the amino acid sequence of SEQ ID NO: 29; and
b. a second polynucleotide encoding a light chain polypeptide having the amino acid sequence of SEQ ID NO: 23.
3. The nucleic acid of claim 2 , wherein the nucleic acid is an expression vector.
4. An expression vector system comprising:
a. a first expression vector comprising a first polynucleotide encoding a heavy chain polypeptide having the amino acid sequence of SEQ ID NO: 29; and
b. a second expression vector comprising a second polynucleotide encoding a light chain polypeptide having the amino acid sequence of SEQ ID NO: 23.
5. A host cell comprising
a. a first polynucleotide encoding a heavy chain polypeptide having the amino acid sequence of SEQ ID NO: 29; and
b. a second polynucleotide encoding a light chain polypeptide having the amino acid sequence of SEQ ID NO: 23.
