Abstract

Described herein are systems relating to a continuous-flow instrument that includes all necessary components for digital droplet quantification without the need to introduce key reagents or collect and transfer droplets between stages of instrument operation. Digital quantification can proceed without any additional fluid or consumable handling and without exposing fluids to risk of external contamination.

Claims (25)

1. A method for conducting an assay on a biological sample, comprising:

(a) providing a first injector comprising a first dispersed phase and a first continuous phase, and a second injector comprising a second dispersed phase and a second continuous phase, wherein said first dispersed phase comprises said biological sample and said second dispersed phase comprises one or more reagents for a reaction;

(b) directing (i) the first dispersed phase from said first injector to a coalescer, and (ii) the second dispersed phase from said second injector to said coalescer, wherein said coalescer coalesces said first dispersed phase and said second dispersed phase to yield a fluid comprising said biological sample;

(c) directing said fluid from said coalescer to a reactor downstream of said coalescer;

(d) in said reactor, subjecting said fluid to conditions sufficient to conduct said reaction on the biological sample; and

(e) using a detector that is in fluid communication with the reactor to detect said biological sample or a derivative thereof.

2. The method of claim 1 , wherein said first injector, said second injector, or both said first injector and said second injector, have a zero dead volume.

3. The method of claim 1 , wherein (b) further comprises using an external source of energy to apply energy to said first dispersed phase, said second dispersed phase, or both said first dispersed phase and said second dispersed phase.

4. The method of claim 3 , wherein said external source of energy provides an electric field between a first electrode and a second electrode positioned on opposing sides of a junction outlet of said coalescer.

5. The method of claim 3 , wherein said energy causes said first dispersed phase and said second dispersed phase to coalesce.

6. The method of claim 1 , further comprising providing a controller.

7. The method of claim 6 , wherein the controller controls a duration of coalescing, a timing of coalescing, or both, of the first dispersed phase and the second dispersed phase.

8. The method of claim 1 , wherein said detector is an optical detector, and wherein (e) comprises detecting an optical signal from said biological sample or derivative thereof.

9. The method of claim 1 , wherein said biological sample comprises a nucleic acid sample.

10. The method of claim 9 , wherein the biological sample comprises DNA, RNA, or both DNA and RNA.

11. The method of claim 1 , wherein the first dispersed phase comprises at least one nucleic acid molecule.

12. The method of claim 9 , wherein the second dispersed phase comprises nucleic acid amplification reagents.

13. The method of claim 12 , wherein (d) comprises using said nucleic acid amplification reagents to conduct said reaction on said nucleic acid sample to yield an amplification product of said nucleic acid sample, and wherein (e) comprises using said detector to detect said amplification product.

14. The method of claim 1 , wherein said reactor comprises a heat source or a thermocycler.

15. The method of claim 1 , wherein (d) comprises conducting said reaction in droplets, and (e) comprises detecting said biological sample or derivative thereof in droplets.

The method of claim 1 , wherein: (a) the first injector is in fluid communication with (i) a reservoir for the first dispersed phase and (ii) a reservoir for the first continuous phase; (b) the second injector is in fluid communication with (i) a reservoir for the second dispersed phase and (ii) a reservoir for the second continuous phase; or both (a) and (b).

17. The method of claim 1 , wherein the first continuous phase, the second continuous phase, or both the first continuous phase and the second continuous phase, comprise an oil.

18. The method of claim 1 , wherein said reaction is a nucleic acid amplification reaction.

19. The method of claim 18 , wherein said nucleic acid amplification reaction is a PCR, digital PCR, or RT-PCR reaction.

20. The method of claim 17 , wherein the oil comprises a fluorinated oil, a silicone oil, a hydrocarbon oil, a mineral oil, or any combination thereof.
