Abstract

A biochip device comprising a substrate constituted by at least one plate of material forming a multimode planar waveguide and carrying chromophore elements suitable for emitting fluorescence in response to excitation by guided waves having an evanescent portion, the device being characterized in that it includes coupling means for coupling excitation light with the waveguide in the form of guided waves, the coupling means being substantially non-directional.

Claims (20)

1. A method of excitation of a biochip device, the method comprising:

receiving and diffusing, by a non-directional diffusing structure of an optical waveguide including a filter layer disposed on a top face of the optical waveguide, the filter layer having a refractive index smaller than the refractive index of the waveguide material, the filter layer being separated from the non-directional diffusing structure, the optical waveguide including an absorption filter disposed on the filter layer, the filter layer being interposed between the top face and the absorption filter, the non-directional diffusing structure being one of: (i) disposed at a first portion of the top face of the optical waveguide, (ii) disposed at a portion of a bottom face of the optical waveguide that is opposite the first portion of the top face of the optical waveguide, and (iii) disposed within the optical waveguide, light incoming from a direction out of a plane of the optical waveguide into the optical waveguide at one of: (i) the first portion of the top face, (ii) the portion of the bottom face opposite the first portion, and (iii) within the optical waveguide to excite a plurality of modes propagating inside the optical waveguide in the plane of the optical waveguide as guided waves through the optical waveguide to a second portion of the top face, the second portion carrying chromophore elements that emit fluorescence when absorbing waveguide excitation light, the non-directional diffusing structure having a disordered spatial distribution of refractive index, the optical waveguide being a plate of material with a thickness in a range supporting multiple light-propagating modes at an excitation wavelength;

filtering, by the filter layer, some of the plurality of light-propagating modes from the optical waveguide and refracting the filtered light-propagating modes inside the filter layer; and

absorbing, by the absorption filter, the filtered light-propagating modes from the optical waveguide so that the filtered light-propagating modes are prevented from reaching the second portion carrying the chromophore elements.

2. The method according to claim 1 , further comprising:

providing a hybridizing chamber disposed on the second portion of the waveguide, the hybridizing chamber configured to house the chromophore elements configured to be excited by the waveguide excitation light; and

extracting guided modes, received at the second portion of the waveguide, for which the effective index is less than or equal to a predetermined threshold value from the plurality of light-propagating modes from the waveguide, the predetermined threshold value corresponding to one or more of a material of the hybridizing chamber and a hybridizing solution within the hybridizing chamber.

3. The method according to claim 1 , further comprising receiving probe molecules at a plurality of pads deposited on the optical waveguide at the second portion, the chromophore elements being configured to hybridize with the probe molecules deposited on the pads.

4. The method according to claim 1 , wherein the non-directional diffusing structure is disposed at the first portion of the top face or the portion of the bottom face of the optical waveguide.

5. The method according to claim 4 , wherein the non-directional diffusing structure comprises a frosted portion.

6. The method according to claim 4 , wherein the non-directional diffusing structure comprises Polytetrafluoroethylene.

7. The method according to claim 4 , wherein the non-directional diffusing structure comprises metallic particles.

8. The method according to claim 4 , wherein the non-directional diffusing structure comprises colloidal particles.

9. The method according to claim 4 , wherein the non-directional diffusing structure comprises diffusing particles in a matrix of a resin.

10. The method according to claim 9 , wherein the resin is one of an acrylic resin, a glycerophthalic resin, and a polymer.

11. The method according to claim 4 , wherein the non-directional diffusing structure comprises microcavities formed inside the waveguide.

The method according to claim 4 , wherein the non-directional diffusing structure is disposed at one of the first portion of the top face of the optical waveguide and the portion of the bottom face of the optical waveguide, the non-directional diffusing structure comprising a layer of fluorophore material.

13. The method according to claim 12 , wherein the fluorophore material comprises quantum dots.

14. The method according to claim 12 , wherein the fluorophore material comprises organic fluorophores.

15. The method according to claim 12 , wherein the fluorophore material comprises fluorophores based on rare earth or luminescent ions.
