Abstract

Methods and apparatus are provided for applying an fragment of a neurotoxin such as the active light chain (LC) of the botulinum toxin (BoNT), such as one of the serotype A, B, C, D, E, F or G botulinum toxins, via permeabilization of targeted cell membranes to enable translocation of the botulinum neurotoxin light chain (BoNT-LC) molecule across the targeted cell membrane to the cell cytosol where a therapeutic response is produced in a mammalian system. The methods and apparatus include use of catheter based delivery systems, non-invasive delivery systems, and transdermal delivery systems.

Claims (34)

1. Apparatus for treating tissue in a nasal cavity, the apparatus comprising:

an elongate body, comprising:

a proximal portion; and

a distal portion configured for positioning in a nasal cavity;

a first electrode comprising a first pair of electrodes and a second electrode comprising a second pair of electrodes, the first electrode and second electrode being spaced away from one another and coupled with the distal portion of the elengate body and configured to deliver energy to a target tissue in the nasal cavity; and

a generator coupled with the proximal portion of the elongate body for supplying energy to at least one of the first electrode or the second electrode to treat the target tissue in the nasal cavity, the generator configured to deliver the energy in pulses between 1 V and 500 V.

2. The apparatus of claim 1 , wherein the first pair of electrodes and the second pair of electrodes each comprise bipolar radiofrequency electrodes.

3. The apparatus of claim 1 , wherein the generator comprises a radiofrequency generator.

4. The apparatus of claim 1 , wherein each of the first pair of electrodes and the second pair of electrodes comprise bipolar radiofrequency electrodes that are movable relative to the distal portion of the elongate body.

5. The apparatus of claim 4 , wherein each of the first pair of electrodes and the second pair of electrodes comprise bipolar radiofrequency electrodes that are configured to deliver energy laterally relative to the distal portion of the elongate body.

6. The apparatus of claim 4 , wherein each of the first pair of electrodes and the second pair of electrodes comprise bipolar radiofrequency electrodes that are configured to penetrate nasal mucosa lining the nasal cavity.

7. The apparatus of claim 1 , wherein each of the first pair of electrodes and the second pair of electrodes comprises a treatment electrode and a stimulating electrode.

8. The apparatus of claim 7 , further comprising a monitoring system operatively coupled to the first electrode and the second electrode for measuring a degree of treatment of the target tissue.

9. Apparatus for treating tissue in a nasal cavity, the apparatus comprising:

an elongate body, comprising:

a proximal portion; and

a distal portion configured for positioning in a nasal cavity;

a first electrode comprising a first pair of electrodes and a second electrode comprising a second pair of electrodes, the first electrode and second electrode being spaced away from one another and coupled with the distal portion of the elongate body and configured to deliver energy to a target tissue in the nasal cavity; and

a generator coupled with the proximal portion of the elongate body for supplying energy to at least one of the first electrode or the second electrode to treat the target tissue in the nasal cavity, the generator configured to deliver the energy in pulses between 1 V and 500 V,

wherein the first electrode and the second electrode are configured to be activated with energy from the generator in a sequential manner.

10. The apparatus of claim 9 , wherein the generator comprises a radiofrequency generator.

11. The apparatus of claim 9 , wherein the first pair of electrodes and the second pair of electrodes each comprise bipolar radiofrequency electrodes.

12. Apparatus for treating tissue in a nasal cavity, the apparatus comprising:

an elongate body, comprising:

a proximal portion, and

a distal portion configured for positioning in a nasal cavity,

a first pair of electrodes and a second pair of electrodes being spaced away from one another at the distal portion of the elongate body and configured to deliver energy to a target tissue in the nasal cavity, and

a generator coupled with the proximal portion of the elongate body for supplying radiofrequency energy to at least one of the first pair of electrodes or the second pair of electrodes to treat the target tissue in the nasal cavity, the generator configured to deliver the radiofrequency energy in pulses between 1 V and 500 V and for a duration between 5 microseconds to 100 milliseconds.

13. The apparatus of claim 1, 9, or 12 , wherein at least the first pair of electrodes comprise bipolar radiofrequency electrodes, wherein the bipolar radiofrequency electrodes are configured to deliver non-reversible poration energy to the target tissue in the nasal cavity.

14. The apparatus of claim 13 , wherein the bipolar radiofrequency electrodes comprise non-penetrating surface electrodes.

15. The apparatus of claim 14 , wherein the bipolar radiofrequency electrodes are configured to deliver the non-reversible poration energy laterally relative to the distal portion of the elongate body.

16. The apparatus of claim 15 , wherein the target tissue in the nasal cavity comprises goblet cells.

17. The apparatus of claim 16 , wherein an outer surface of the bipolar radiofrequency electrodes is planar with an outer surface of the elongate body.

18. The apparatus of claim 17 , wherein the elongate body defines a portion of a flexible catheter, wherein the flexible catheter comprises a sheath for housing the elongate body, wherein the distal portion of the elongate body is steerable.
