Abstract

A microstructured hemostat comprising multiple layers of microstructure, each layer characterized by one or more length scales, is described. Microstructured hemostats of the present invention, can reduce the time for blood coagulation, control the morphology of the coagulation, and provide a novel diagnostic platform for evaluation of coagulation function from a morphological perspective.

Claims (20)

1. A device for promoting blood coagulation comprising:

a hierarchically arranged microstructured surface comprising at least a first microstructure and a second microstructure, the second microstructure disposed about the first microstructure, wherein the first microstructure and second microstructure are configured to have a fractal dimension of between 2 and 3, the first microstructure and second microstructure configured to create a plurality of spatially distributed surface energy gradients via Wenzel-Cassie wetting states, wherein the first microstructure or the second microstructure comprises fluted or finned side structures and the fluted or finned side structures are interpenetrating, and wherein the microstructured surface is configured to promote thrombus formation when in contact with blood.

2. The device of claim 1 wherein the first microstructure has a pitch of 1000 microns and the second microstructure has a pitch of 100 microns.

3. The device of claim 1 wherein the microstructured surface has a branching ratio wherein the height of the first microstructure is ten times the height of the second microstructure.

4. The device of claim 1 wherein the first microstructure and the second microstructure are arranged in a triangular grid pattern, wherein the first microstructure has a height of about 400 microns and the second microstructure has a height of about 45 microns.

5. The device of claim 1 wherein the fractal dimension is between 2.06 and 2.08.

6. The device of claim 1 wherein the microstructured surface is configured to have an activated partial thromboplastin time that is reduced relative to a surface that lacks a hierarchical microstructure arrangement.

7. The device of claim 1 wherein the microstructured surface further comprises a chemical coagulant, the chemical coagulant being cross-linked to the first microstructure and second microstructure.

8. The device of claim 7 wherein the chemical coagulant comprises Gardenia fruit extract.

9. The device of claim 7 wherein the chemical coagulant comprises free amino groups of collagen, gelatin, or both, and wherein the microstructured surface further comprises a polymer substrate containing at least one aldehyde group.

10. The device of claim 7 wherein the chemical coagulant comprises at least one oxidized polysaccharide.

11. A device for promoting blood coagulation comprising:

a hierarchically arranged microstructured surface comprising at least a first microstructure, a second microstructure, and a third microstructure surface, the second microstructure disposed about the first microstructure and the third microstructure disposed about the second microstructure, wherein the first microstructure and second microstructure are configured to have a fractal dimension of between 2 and 3, the first microstructure, second microstructure, and third microstructure configured to create a plurality of spatially distributed surface energy gradients via Wenzel-Cassie wetting states, wherein the first microstructure or the second microstructure comprises fluted or finned side structures and the fluted or finned side structures are interpenetrating, and wherein the microstructured surface is configured to promote thrombus formation when in contact with blood.

12. The device of claim 11 wherein the first microstructure has a pitch of 1000 microns, the second microstructure has a pitch of 100 microns, and the third microstructure has a pitch of 10 microns.

13. The device of claim 11 wherein the microstructured surface has a branching ratio wherein the height of the first microstructure is ten times the height of the second microstructure, and the second microstructure is ten times the height of the third microstructure.

The device of claim 11 wherein the first microstructure, the second microstructure, and the third microstructure are arranged in a triangular grid pattern, wherein the first microstructure has a height of about 400 microns, the second microstructure has a height of about 45 microns, and the third microstructure has a height of about 15 microns.

15. The device of claim 11 wherein the microstructured surface further comprises a chemical coagulant, the chemical coagulant being cross-linked to the first microstructure, second microstructure, and third microstructure.

16. The device of claim 15 wherein the chemical coagulant comprises Gardenia fruit extract.

17. The device of claim 15 wherein the chemical coagulant comprises free amino groups of collagen, gelatin, or both, and wherein the microstructured surface further comprises a polymer substrate containing at least one aldehyde group.

18. The device of claim 15 wherein the chemical coagulant comprises at least one oxidized polysaccharide.
