Abstract

A full-field microscopy method for detection of Brillouin-scattered light includes illuminating a two-dimensional plane in a sample with interrogating light having a first wavelength. Light emitted from the two-dimensional plane can be collected. The emitted light comprises Brillouin-scattered light resulting from interaction of the interrogating light with the sample. The Brillouin-scattered light can have a second wavelength shifted from the first wavelength. The collected light can be passed through a spectrally-selective assembly comprising a gas or vapor illuminated by pumping light. After the spectrally-selective assembly, the Brillouin-scattered light from multiple points in the two-dimensional plane in the sample can be simultaneously detected by an electro-optical sensor. In some embodiments, the spectrally-selective assembly can be altered by changing a wavelength or polarization of the pumping light to allow acquisition of a Brillouin spectrum.

Claims (38)

1 . A method comprising:

simultaneously illuminating multiple points in a sample with interrogating light having a first wavelength;

illuminating an atomic vapor cell by pumping light; and

selecting, via a detection arm, for Brillouin-scattered light in light simultaneously emitted from the multiple simultaneously-illuminated points in the sample, the Brillouin-scattered light having a second wavelength shifted from the first wavelength, the detection arm comprising the illuminated atomic vapor cell,

wherein the atomic vapor cell changes a polarization of the Brillouin-scattered light passing therethrough that has the second wavelength, and

the detection arm includes one or more polarization optical elements that select for the Brillouin-scattered light using the changed polarization.

2 . The method of claim 1 , wherein an input optical axis for the detection arm is parallel to an optical axis of the interrogating light.

3 . The method of claim 1 , wherein an input optical axis for the detection arm is at an angle of about 96° with respect to an optical axis of the interrogating light.

4 . The method of claim 1 , wherein one or more optical properties of the atomic vapor cell are changed by illumination with the pumping light.

5 . The method of claim 4 , wherein the one or more optical properties include absorption, dichroism, polarization, or birefringence.

6 . The method of claim 5 , wherein the illumination with the pumping light changes dichroism properties for wavelengths corresponding to a ground state or excited state of gas or vapor in the atomic vapor cell.

7 . The method of claim 1 , further comprising:

detecting, using an electro-optical sensor, the Brillouin-scattered light selected via the detection arm,

wherein the Brillouin-scattered light from the multiple points in the sample is simultaneously detected by the electro-optical sensor.

8 . The method of claim 7 , wherein the detection arm is configured to allow a fixed band of wavelengths to pass, and the method further comprises associating an intensity at each pixel of the electro-optical sensor to a shift of a Brillouin peak.

9 . The method of claim 1 , wherein the interrogating light is in a form of a light sheet.

10 . A method comprising:

simultaneously illuminating multiple points in a sample with interrogating light having a first wavelength;

selecting, via a detection arm, for Brillouin-scattered light in light simultaneously emitted from the multiple simultaneously-illuminated points in the sample, the Brillouin-scattered light having a second wavelength shifted from the first wavelength, the detection arm including an atomic vapor cell illuminated by pumping light;

altering the atomic vapor cell by saturating a different electron transition of gas or vapor therein;

after the altering, simultaneously illuminating the multiple points in the sample with the interrogating light having the first wavelength; and

selecting, via the detection arm with the altered atomic vapor cell, for other Brillouin-scattered light in light simultaneously emitted from the multiple simultaneously-illuminated points in the sample, the other Brillouin-scattered light having a third wavelength different from the first and second wavelengths.

11 . The method of claim 10 , wherein the altering comprises changing a wavelength of the pumping light, a polarization of the pumping light, or both the wavelength and polarization of the pumping light.

12 . The method of claim 10 , wherein the altering comprises applying an external magnetic field to the atomic vapor cell.

13 . A system comprising:

a Brillouin microscopy modality comprising:

a first light source providing interrogating light having a first wavelength;

an illumination optical assembly constructed to simultaneously illuminate multiple points in a sample with the interrogating light;

a pump light source providing pumping light; and

a detection optical assembly comprising an atomic vapor cell illuminated by the pumping light, the detection optical assembly being constructed to select for Brillouin-scattered light in light simultaneously emitted from the multiple simultaneously-illuminated points in the sample, the Brillouin-scattered light having a second wavelength shifted from the first wavelength,

wherein the detection optical assembly comprises one or more polarization optical elements,

the atomic vapor cell is constructed to change a polarization of the Brillouin-scattered light passing therethrough that has the second wavelength, and

the one or more polarization optical elements use the changed polarization to select for the Brillouin-scattered light.

14 . The system of claim 13 , wherein an input optical axis for light from sample to the detection optical assembly is parallel to an optical axis of the interrogating light from the illumination optical assembly to the sample.

15 . The system of claim 13 , wherein an input optical axis for light from the sample to the detection optical assembly is at an angle of about 96° with respect to an optical axis of the interrogating light from the illumination optical assembly to the sample.

16 . The system of claim 13 , further comprising a magnetic device constructed to apply a transition-altering magnetic field to the atomic vapor cell.

17 . The system of claim 13 , further comprising an electro-optical sensor constructed to detect the Brillouin-scattered light selected by the detection optical assembly.

18 . The system of claim 13 , further comprising a non-Brillouin microscopy modality employing an optical imaging or spectroscopy technique.
