Abstract

Disclosed herein are compositions comprising containers and silk elements. Disclosed herein are methods of regenerating an at least partially severed nerve cell. Disclosed herein are compositions for regenerating an at least partially severed nerve cell.

Claims (31)

1 . A method comprising:

a) providing a first plurality of silk elements and a second plurality of silk elements, wherein at least one individual silk element in each plurality of silk elements is at least partially individually coated with a first hydrophilic coating;

b) at least partially coating with a second hydrophilic coating:

(i) said first plurality of silk elements to create a first bundle; and

(ii) said second plurality of silk elements to create a second bundle;

c) bundling at least said first bundle and said second bundle into a bundle of bundles, and at least partially coating said bundle of bundles with a third hydrophilic coating;

d) bundling at least two of said bundle of bundles into a bundle of bundles of bundles, wherein said bundle of bundles of bundles is at least partially coated with a fourth hydrophilic coating; and

e) placing said bundle of bundles of bundles at least partially within a lumen of a sheath.

The method of claim 1 , wherein said sheath comprises a protein, a collagen, a gelatin, a silicone, a polymer, a polyester, a hydrophilic material, a polyol, a hybrid composition, an isolated tissue, an isolated tissue product, a decellularized nerve conduit, a carbohydrate, a biomimetic material, a silk, a silk composite, or any combination thereof.

3 . The method of claim 1 , wherein said sheath comprises an isolated tissue product.

4 . The method of claim 1 , wherein said sheath comprises a collagen.

5 . The method of claim 1 , wherein said sheath comprises a polyglycolic acid (PGA).

6 . The method of claim 1 , wherein at least one silk element from either said first bundle or said second bundle spans a majority of a length of said sheath after placing each of said first bundle and said second bundle at least partially within said lumen of said sheath.

7 . The method of claim 1 , wherein said first hydrophilic coating comprises an at least partial coating of hyaluronic acid.

8 . The method of claim 1 , wherein said second hydrophilic coating comprises an at least partial coating of hyaluronic acid.

9 . The method of claim 1 , further comprising treating a nerve injury by implanting a nerve conduit into a subject, wherein said nerve conduit comprises said bundle of bundles of bundles at least partially within said lumen of said sheath.

10 . The method of claim 1 , wherein said sheath comprises a tube.

11 . The method of claim 10 , wherein said tube comprises a proximal end and a distal end, wherein at least part of said tube comprises an additional constituent, wherein said additional constituent is distributed in a gradient from said proximal end to said distal end, and wherein said additional constituent encourages a growth of an axon.

12 . The method of claim 10 , wherein said tube comprises a plurality of pores.

13 . The method of claim 1 , wherein said at least partially coating with said second hydrophilic coating comprises submerging each of said first bundle and said second bundle in a hydrophilic substance individually, wherein said second hydrophilic coating comprises said hydrophilic substance.

14 . A method comprising placing a bundle of bundles of bundles of silk elements into a lumen of a sheath, wherein said bundle of bundles of bundles comprises at least two bundles of bundles, wherein each of said at least two bundles of bundles comprises at least two bundles of silk elements; wherein:

a) each of said at least two bundles of silk elements comprise a plurality of silk elements, wherein each individual silk element in each of said plurality of silk elements comprises a first hydrophilic coating at least partially around said individual silk element;

b) each of said at least two bundles of silk elements comprises a second hydrophilic coating around each of said at least two bundles of silk elements which holds in place each of said at least two bundles of silk elements;

c) each of said at least two bundles of bundles comprises a third hydrophilic coating around each of said at least two bundles of bundles which holds in place each of said at least two bundles of bundles; and

d) said bundle of bundles of bundles comprises a fourth hydrophilic coating around said bundle of bundles of bundles which holds in place said bundle of bundles of bundles.

The method of claim 14 , wherein said sheath comprises a protein, a collagen, a gelatin, a silicone, a polymer, a polyester, a hydrophilic material, a polyol, a hybrid composition, an isolated tissue, an isolated tissue product, a decellularized nerve conduit, a carbohydrate, a biomimetic material, a silk, a silk composite, or any combination thereof.

16 . The method of claim 14 , wherein said sheath comprises an isolated tissue product.

17 . The method of claim 14 , wherein said sheath comprises a collagen.

18 . The method of claim 14 , wherein said sheath comprises a polyglycolic acid (PGA).

19 . The method of claim 14 , wherein said sheath comprises a tube.

20 . The method of claim 19 , wherein said tube comprises a plurality of pores.
