Abstract

Methods of preparing multi-phase soft gelatin dosage forms comprising at least one preformed solid dosage form comprising a statin compound and at least one liquid fill phase comprising Omega-3 fatty acids. The multi-phase soft gelatin dosage forms combine at least one solid dosage form and at least one liquid phase for single ingestion. The solid phase, liquid phase and/or coatings may include additional active pharmaceutical ingredients, nutraceuticals, nutritional supplements, or therapeutic substances, functional excipients or combinations thereof.

Claims (28)

1 . A method of preparing a multi phase soft gelatin capsule for oral administration, the method comprising:

introducing at least one preformed tablet comprising a statin compound into a first soft capsule shell part;

independently introducing at least one liquid fill phase comprising omega-3 fatty acids into the first soft capsule shell part, wherein the at least one preformed tablet is within the at least one liquid fill phase and the at least one liquid fill phase does not comprise a capsule; and

sealing the first soft capsule shell part and a soft second soft capsule shell part together to form the multi phase soft gelatin capsule.

2 . The method of claim 1 , wherein the at least one preformed tablet is a pharmaceutical grade finished dosage form.

3 . The method of claim 1 , wherein the at least one liquid fill phase further comprises at least one additional active ingredient.

4 . The method of claim 3 , wherein the at least one additional active ingredient comprises an active pharmaceutical ingredient, nutraceutical, nutritional supplement, therapeutic substance, functional excipients or combinations thereof.

5 . The method of claim 1 , wherein the at least one liquid fill phase further comprises one or more diluent oils.

6 . The method of claim 5 , wherein the one or more diluent oils comprise vegetable oil, mineral oil, food grade oil, pharmaceutically acceptable oil or mixtures thereof.

7 . The method of claim 1 , wherein the statin compound comprises atorvastatin, fluvastatin, lovastatin, pitavastatin, pravastatin, rosuvastatin, simvastatin, a pharmaceutically acceptable salt thereof, or combinations thereof.

8 . The method of claim 1 , wherein the statin compound comprises rosuvastatin, a pharmaceutically acceptable salt thereof, or a combination thereof, and wherein the liquid fll phase comprises one or more omega-3 fatty acids.

9 . The method of claim 1 , wherein the statin compound further comprises at least one additional active ingredient.

10 . The method of claim 9 , wherein the at least one additional active ingredient is an active pharmaceutical ingredient.

11 . The method of claim 9 , wherein:

the statin compound is simvastatin and the at least one additional active ingredient is exetimibe;

the statin compound is lovastatin and the at least one additional active ingredient is niacin;

the statin compound is atorvastatin calcium and the at least one additional active ingredient is amlodipine besylate; or

the statin compound is simvastatin and the at least one additional active ingredient is niacin.

12 . The method of claim 1 , wherein the at least one preformed tablet has at least one coating.

13 . The method of claim 12 , wherein the at least one coating comprises an immediate release coating, protective coating, enteric coating, delayed release coating, sustained release coating, barrier coating, moisture shield coating, or combinations thereof.

14 . The method of claim 12 , wherein the at least one coating on the at least one preformed tablet is selected from the group consisting of: film coating, gelatin coating, and combinations thereof.

15 . The method of claim 12 , wherein the at least one coating on the at least one preformed tablet further comprises an active pharmaceutical ingredient, nutraceutical, nutritional supplement, therapeutic substance, functional excipients, or combinations thereof.

16 . The method of claim 1 , wherein the omega-3 fatty acids comprise triglycerides, ethyl esters or an emulsified formulation of the omega-3 fatty acids.

17 . The method of claim 16 , wherein the omega-3 fatty acids comprise eicosapentaenoic acid (EPA), docosahexaenoic acid (DHA), or a mixture thereof.

18 . The method of claim 1 , wherein the soft gelatin capsule comprises at least one coating.

19 . The method of claim 18 , wherein the at least one coating comprises an immediate release coating, protective coating, enteric coating, delayed release coating, sustained release coating, barrier coating, or combinations thereof.

20 . The method of claim 18 , wherein the at least one coating on the soft gelatin capsule further comprises an active pharmaceutical ingredient, nutraceutical, nutritional supplement, therapeutic substance, functional excipient, or combinations thereof.

21 . A method of treating at least one of hypercholesterolemia or hyperlipidemia in a subject, comprising: administering to the subject a multi phase soft gelatin capsule prepared by the method of claim 1 .
