Abstract

A portable diagnostic device has a lysate stage ( 167 ) with a port for receiving a sample and containing magnetic beads with a probe, and an outlet port. A series of assay stages ( 161 - 164 ) are linked with the lysate vessel, each with a reservoir linked by channels. The final stage ( 164 ) has a sensor ( 169 ) for detecting beads attached to analyte molecules which have been conveyed according to attachment to probes on beads. Larger transport beads cause reporter beads which are tethered by target NA and probes to be transported to the final sensor stage, where they are released and detected when the transport beads have been removed.

Claims (28)

1 . A diagnostic system comprising a microfluidic cartridge device and a reader:

wherein the device comprises an inlet for receiving a sample comprising a target analyte and at least one assay stage comprising a sensor, wherein said sensor comprises a sensing region and a plurality of probes attached to the sensing region and an integrated circuit comprising a plurality of electrodes disposed below a surface of the sensing region, wherein said probes are configured to capture the target analyte from the sample and the electrodes are configured to measure a change in capacitance of the sensing region using fringe-field sensing upon capturing the target analyte;

wherein the reader is configured to receive the device and comprises a means for communication with and control of the sensor of the device, and wherein the sensor comprises a CMOS integrated circuit chip, and wherein the device comprises an elastomer strip for contacting pins of the sensor.

2 . The system of claim 1 , wherein the device comprises:

a housing;

one or more assay stages, each with a reservoir linked by channels; and

wherein at least one of the one or more assay stages includes the sensor for detecting the target analyte which is attached to reporter beads (R-Beads); and

wherein the reader comprises:

a support for receiving the device;

movable magnets to convey magnetic transport beads (T-Beads) in the device through the assay stages of the device,

a heat source,

wherein the sensor is arranged to read the R-beads on its surface.

3 . The system of claim 2 , wherein the magnets walk the T-beads through the one or more assay stages to perform steps of enrichment, purification, washing, tethering, and sensing.

4 . The system of claim 3 , wherein the T-beads are coated with PNA, and the target analyte is DNA or RNA which hybridizes to the PNA probes on the beads.

5 . The system of claim 4 , wherein the magnets have a drive to vary separation of the magnets from the device, for analyte mixing.

6 . The system of claim 5 , wherein the assay stages and the inlet comprise a plurality of different probes on the T-beads beads and arranged to target multiple nucleic acid target analytes from the sample.

7 . The system of claim 6 , wherein the T-beads with different targets arrive on the sensor and the sensor determines the quantities of each type of R-bead.

8 . The system of claim 7 , wherein the sensor reads the R-beads on its surface against a background of an organic liquid having a low dielectric constant.

9 . The system of claim 8 , wherein the first assay stage is a circular lysis chamber which slots into a butterfly needle safety-tube holder to receive a venipuncture whole blood sample.

10 . The system of claim 9 , wherein cell-free viral RNA in the whole blood sample is distinguished from viral DNA by an in-built plasma separation filter and/or by the higher melt temperature of PNA-RNA compared to PNA-DNA.

The system of claim 10 , wherein the cell-free viral RNA from the sample captured on the T-beads is bound to a second PNA probe on a R-bead in an assay reservoir stage, forming a transport bead-reporter bead tethered sandwich in which the R-beads have a 1:1 ratio to the target analyte cell-free viral RNA.

12 . The system of claim 11 , wherein the sensor quantifies the R-beads, thereby quantifying the cell-free viral RNA in the sample.

13 . The system of claim 1 , wherein the CMOS IC comprises EEPROM storage for a unique sample identifier.

14 . The system of claim 1 , wherein the integrated circuit chip has converters and digital calibration circuits which enable self-testing and calibration of the amount of probe on each sensor.

15 . The system of claim 14 , wherein the integrated circuit chip comprises a an on-chip non-volatile memory.

16 . The system of claim 15 , wherein the non-volatile memory records the self-testing and/or calibration.

17 . The system of claim 16 , wherein the system is configured to perform self-testing and calibration during wafer-level testing.

The system of claim 17 , wherein the integrated circuit comprises a temperature sensor operable to determine the temperature of the device, wherein when the device is inserted into the reader unit, a heater in the reader brings the device to a start temperature and a temperature sensor provides feedback from the temperature sensor to the reader.
