Abstract

The present invention relates to methods of using compounds of Formula I, wherein R 1a , R 1b , R 2 , R 3 , R 4 , W, Y, and Z are as described herein, and pharmaceutically acceptable salts thereof. The compounds are potentiators of Cystic Fibrosis Transmembrane conductance Regulator (CFTR). The invention also discloses pharmaceutical compositions comprising the compound, optionally in combination with additional therapeutic agents, and methods of potentiating, in mammals, including humans, CFTR by administration of the compounds. These compounds are useful for the treatment of cystic fibrosis (CF), asthma, bronchiectasis, chronic obstructive pulmonary disease (COPD), constipation, Diabetes mellitus, dry eye disease, pancreatitis, rhinosinusitis, Sjögren's Syndrome, and other CFTR associated disorders.

Claims (135)

1 . A pharmaceutical composition comprising a therapeutically effective amount of one or more of a compound of Formula I or a pharmaceutically acceptable salt thereof, one or more additional therapeutic agents, and a pharmaceutically acceptable carrier,

wherein

W is selected from the group consisting of phenyl, which is optionally fused with a five to six membered cycloalkyl or a five to six membered heterocycloalkyl comprising one, two, three, or four heteroatoms selected independently for each occurrence from the group consisting of N, O and S(O) n ;

a five to ten membered heteroaryl comprising one, two, three, or four heteroatoms selected independently for each occurrence from the group consisting of N, O and S(O) n ;

a four to seven membered heterocycloalkyl comprising one, two, three, or four heteroatoms selected independently for each occurrence from the group consisting of N, O and S(O) n ; and

C 3 -C 7 cycloalkyl;

wherein the phenyl, five to ten membered heteroaryl, four to seven membered heterocycloalkyl, and C 3 -C 7 cycloalkyl are optionally substituted with one, two, three, four, or five R 5 ;

Y is a five membered heteroaryl comprising one, two, three, or four heteroatoms selected independently for each occurrence from the group consisting of N, O and S(O) n , wherein the five membered heteroaryl is optionally substituted with one, two, or three substituents independently selected from the group consisting of halo, C 1 -C 6 alkyl, and C 1 -C 6 haloalkyl;

Z is selected from the group consisting of phenyl, C 1 -C 6 alkyl, C 3 -C 7 cycloalkyl, a five or six membered heteroaryl comprising one, two or three heteroatoms selected independently for each occurrence from the group consisting of N, O and S(O) n , and a four to seven membered heterocycloalkyl comprising one, two, or three heteroatoms selected independently for each occurrence from the group consisting of N, O and S(O) n ; wherein the phenyl, C 1 -C 6 alkyl, C 3 -C 7 cycloalkyl, five or six membered heteroaryl, and four to seven membered heterocycloalkyl are each independently optionally substituted with one, two, three, four, or five R 6 ;

R 1a and R 1b are each independently selected from the group consisting of —H, —OH, halo, C 1 -C 6 alkyl, C 3 -C 7 cycloalkyl, and a four to seven membered heterocycloalkyl comprising one, two, or three heteroatoms each independently selected from the group consisting of N, O and S(O) n , wherein the C 1 -C 6 alkyl group is optionally substituted with one, two, or three substituents each independently selected from the group consisting of halo, —OH, C 1 -C 3 alkyoxy, C 3 -C 7 cycloalkyl, and a four to seven membered heterocycloalkyl comprising one, two, or three heteroatoms each independently selected from the group consisting of N, O and S(O) n ; and wherein each C 3 -C 7 cycloalkyl and each four to seven membered heterocycloalkyl are optionally substituted with one, two or three substituents independently selected for each occurrence from the group consisting of —OH, halo, and C 1 -C 6 alkyl;

or R 1a and R 1b taken together with the carbon to which they are attached form a C 3 -C 7 cycloalkyl or a four to seven membered heterocycloalkyl comprising one, two, or three heteroatoms each independently selected from the group consisting of N, O and S(O) n ; and wherein the C 3 -C 7 cycloalkyl and the four to seven membered heterocycloalkyl are each optionally substituted with one, two or three substituents each independently selected from the group consisting of —OH, halo, and C 1 -C 6 alkyl;

R 2 is selected from the group consisting of —H, halo, —CN, C 1 -C 6 alkyl and C 1 -C 6 haloalkyl;

R 3 and R 4 are independently selected for each occurrence from the group consisting of —H, C 1 -C 6 alkyl and C 1 -C 6 haloalkyl;

R 5 at each occurrence is independently selected from the group consisting of halo, —CN, C 1 -C 6 alkyl, C 1 -C 6 haloalkyl, —OR 7 , —N(R 7 ) 2 , —N(R 7 )C(O)R 7 , —SR 7 , oxo, C 2 -C 7 alkoxyalkyl, —S(O) 2 C 1 -C 6 alkyl, —C(O)R 7 , and a five membered heteroaryl comprising one, two, three, or four heteroatoms selected independently for each occurrence from the group consisting of N, O and S(O) n , wherein the five membered heteroaryl is optionally substituted with one, two, or three substituents independently selected for each occurrence from the group consisting of halo, —CN, C 1 -C 6 alkyl, C 1 -C 6 haloalkyl, —OR 7 , —N(R 7 ) 2 and —SR 7 ;

R 6 at each occurrence is independently selected from the group consisting of halo, C 1 -C 6 alkyl and C 1 -C 6 haloalkyl, —OR 7 , —N(R 7 ) 2 and —SR 7 ;

R 7 is independently selected for each occurrence from the group consisting of —H, C 1 -C 6 alkyl, C 1 -C 6 haloalkyl, C 3 -C 7 cycloalkyl, and C 1 -C 6 alkylC 3 -C 7 cycloalkyl; and

n at each occurrence is independently 0, 1, or 2, or a pharmaceutically acceptable salt thereof together with a pharmaceutically acceptable carrier.

2 . The pharmaceutical composition of claim 1 , wherein R 3 and R 4 are both —H.

3 . The pharmaceutical composition of claim 2 , wherein Y is selected from the group consisting of pyrazolyl, triazolyl, imidazolyl, and isoxazolyl, each of which is optionally substituted with a C 1 -C 6 alkyl.

4 . The pharmaceutical composition of claim 3 , wherein W is phenyl, optionally substituted with one, two, or three R 5 .

5 . The pharmaceutical composition of claim 3 , wherein W is selected from the group consisting of pyrimidinyl, pyridinyl, pyrazinyl, and pyrazolyl, each optionally substituted with one, two, or three R 5 .

6 . The pharmaceutical composition of claim 5 , wherein W is

 optionally substituted with one, two, or three R 5 ;

R 5 at each occurrence is independently selected from the group consisting of —OCH 3 , —CHF 2 , —CF 3 , and —N(CH 3 ) 2 .

7 . The pharmaceutical composition of claim 3 , wherein W is C 3 -C 7 cycloalkyl, optionally substituted with one, two, or three R 5 .

8 . The pharmaceutical composition of claim 3 , wherein Z is selected from the group consisting of phenyl, C 3 -C 7 cycloalkyl, and C 1 -C 6 alkyl, each optionally substituted with one, two, or three R 6 .

9 . The pharmaceutical composition of claim 2 , wherein the moiety Y—Z is selected from the group consisting of:

10 . The pharmaceutical composition of claim 2 , wherein R 2 is selected from the group consisting of: —H, —CN, and —Br.

11 . The pharmaceutical composition of claim 1 , wherein the compound is selected from the group consisting of

4-amino-5-[2-(difluoromethyl)pyrimidin-5-yl]-7-{(1S)-1-[1-(2-fluorophenyl)-1H-pyrazol-4-yl]ethyl}-7H-pyrrolo[2,3-d]pyrimidine-6-carbonitrile;

4-amino-5-[2-(difluoromethyl)pyrimidin-5-yl]-7-{(1R)-1-[1-(2-fluorophenyl)-1H-pyrazol-4-yl]ethyl}-7H-pyrrolo [2,3-d]pyrimidine-6-carbonitrile;

4-amino-7-{(1S)-1-[1-(2-fluorophenyl)-1H-pyrazol-4-yl]propyl}-5-[2-(trifluoromethyl)pyrimidin-5-yl]-7H-pyrrolo[2,3-d]pyrimidine-6-carbonitrile;

The pharmaceutical composition of claim 1 , wherein the one or more additional therapeutic agents are independently selected from the group consisting of a CFTR potentiator, a CFTR corrector, an epithelial sodium channel (ENaC) inhibitor, a CFTR amplifier, a CFTR stabilizer, a read-through agent, an oligonucleotide patch, an autophagy inducer, and a proteostasis modulator.

13 . The pharmaceutical composition of claim 12 , wherein the CFTR potentiator at each occurrence is selected from the group consisting of VX-770 (Ivacaftor), GLPG-1837, GLPG-2451, QBW-251, GLPG-3067, FDL-176, FDL-129, CTP-656, and PTI-P271.

14 . The pharmaceutical composition of claim 12 , wherein the CFTR corrector at each occurrence is selected from the group consisting of VX-809 (lumacaftor), VX-661 (tezacaftor), VX-983, VX-152, VX-440, VX-659, GLPG-2737, P247-A, FDL-304, NU-001, NU-002, GLPG-2222, GLPG-2665, GLPG-2851, FDL-169, and PTI-C1811.

15 . The pharmaceutical composition of claim 12 , wherein the epithelial sodium channel (ENaC) inhibitor at each occurrence is selected from the group consisting of SPX-101, QBW-276 and VX-371.

16 . The pharmaceutical composition of claim 12 , wherein the CFTR amplifier at each occurrence is selected from the group consisting of PTI-428 and PTI-130.

17 . The pharmaceutical composition of claim 12 , wherein the CFTR stabilizer is N-91115 (Cavosonstat).

18 . The pharmaceutical composition of claim 12 , wherein the read-through agent is ataluren.

19 . The pharmaceutical composition of claim 12 , wherein the oligonucleotide patch is QR-010.

20 . The pharmaceutical composition of claim 12 , wherein the autophagy inducer at each occurrence is selected from the group consisting of CX-4945 and the combination of cysteamine and epigallocatechin gallate (EGCG).
