Abstract

The present invention is related to the field of therapeutic delivery platforms. Such therapeutic delivery platforms are based upon the formation of proteinacous exovesicles that bind nucleic acid payloads (e.g., mRNA, siRNA, shRNA etc.). These nucleic acid payloads may hind to the proteinacous vesicle either directly or as an adduct of a linker molecule. As one example, the linker molecule may be an arc 3′UTR that is bound to an Arc protein exovesicle.

Claims (10)

1 . An extracellular vesicle comprising:

i) an encapsulated Drosophila activity-regulated cytoskeleton 1 (darc1) protein;

ii) a nucleic acid comprising a fragment of a darc1 messenger ribonucleic acid 3′ untranslated region (darc1 mRNA 3′ UTR) that is the sequence of SEQ ID NO: 2 or SEQ ID NO 3 and bound to said darc1 protein; and

iii) a non-arc nucleic acid payload fused to said nucleic acid of part (ii).

2 . The extracellular vesicle of claim 1 , wherein said non-arc nucleic acid payload is a deoxyribonucleic acid sequence or a ribonucleic acid sequence.

3 . The extracellular vesicle of claim 1 , wherein said compound is non-arc nucleic acid payload encodes a protein.

4 . The extracellular vesicle of claim 1 , wherein said non-arc nucleic acid payload is selected from the group consisting of an siRNA, an shRNA and an RNAi.

5 . The extracellular vesicle of claim 1 , wherein said extracellular vesicle is a nucleic acid delivery platform.

6 . The extracellular vesicle of claim 1 , wherein said extracellular vesicle is an arc protein capsid.

7 . The extracellular vesicle of claim 1 , wherein said nucleic acid payload further encodes an actin promoter.
