Abstract

Provided herein are various embodiments relating to antibodies and fusion proteins and uses thereof. Some of the embodiments include antibodies that bind CCR8. Some of the embodiments include fusion proteins that bind CCR8. Such antibodies and fusion proteins can be used in methods to treat, for example, cancer.

Claims (38)

1 . An isolated antibody that binds human CCR8, wherein the antibody comprises:

a heavy chain complementarity determining region 1 (HCDR1) comprising the amino acid sequence of SEQ ID NO: 84, a heavy chain complementarity determining region 2 (HCDR2) comprising the amino acid sequence of SEQ ID NO: 85, a heavy chain complementarity determining region 3 (HCDR3) comprising the amino acid sequence of SEQ ID NO: 86, a light chain complementarity determining region 1 (LCDR1) comprising the amino acid sequence of SEQ ID NO: 87, a light chain complementarity determining region 2 (LCDR2) comprising the amino acid sequence of SEQ ID NO: 88, and a light chain complementarity determining region 3 (LCDR3) comprising the amino acid sequence of SEQ ID NO: 89;

wherein the antibody comprises a heavy chain comprising an amino acid sequence having at least 98% sequence identity to SEQ ID NO: 94.

2 . The isolated antibody of claim 1 , wherein the antibody comprises a variable heavy chain comprising an amino acid sequence of SEQ ID NO: 92, and/or a variable light chain comprising the amino acid sequence of SEQ ID NO: 93.

3 . The isolated antibody of claim 1 , wherein the heavy chain comprises an amino acid sequence having at least 99% sequence identity to SEQ ID NO: 94.

4 . The isolated antibody of claim 3 , wherein the heavy chain comprises the amino acid sequence of SEQ ID NO: 94.

5 . The isolated antibody of claim 1 , wherein the heavy chain comprises one to three constant region mutations.

6 . The isolated antibody of claim 5 , wherein the heavy chain comprises one constant region mutation.

7 . The isolated antibody of claim 1 , wherein the antibody comprises a light chain comprising the amino acid sequence of SEQ ID NO: 95.

8 . The isolated antibody of claim 6 , wherein the one to three constant region mutations enhance cell killing.

9 . The isolated antibody of claim 8 , wherein the enhanced cell killing is enhanced antibody-dependent cellular cytotoxicity (ADCC) and/or complement-dependent cytotoxicity (CDC).

10 . The isolated antibody of claim 9 , wherein the one to three constant region mutations are selected from L234, L235, G236, S239, F243, H268, D270, R292, S298, Y300, V305, K326, A330, 1332, E333, K334, and P396.

11 . The isolated antibody of claim 10 , wherein one to three constant region mutations are selected from S239D, S239M, F243L, H268D, D270E, R292P, S298A, Y300L, V305I, K326D, A330L, A330M, 1332E, E333A, K334A, K334E, and P396L.

12 . The isolated antibody of claim 10 , wherein the one to three constant region mutations are selected from: S239D/1332E, S239D/1332E/A330L, and S298A/E333A/K334A.

13 . The isolated antibody of claim 1 , wherein the antibody is afucosylated and/or galactosylated.

14 . An isolated antibody that binds human CCR8, wherein the antibody comprises:

a heavy chain complementarity determining region 1 (HCDR1) comprising the amino acid sequence of SEQ ID NO: 100, a heavy chain complementarity determining region 2 (HCDR2) comprising the amino acid sequence of SEQ ID NO: 85, a heavy chain complementarity determining region 3 (HCDR3) comprising the amino acid sequence of SEQ ID NO: 86, a light chain complementarity determining region 1 (LCDR1) comprising the amino acid sequence of SEQ ID NO: 87, a light chain complementarity determining region 2 (LCDR2) comprising the amino acid sequence of SEQ ID NO: 88, and a light chain complementarity determining region 3 (LCDR3) comprising the amino acid sequence of SEQ ID NO: 89;

wherein the antibody comprises a heavy chain comprising an amino acid sequence having at least 98% sequence identity to SEQ ID NO: 98.

15 . The isolated antibody of claim 14 , wherein the antibody comprises a variable heavy chain comprising an amino acid sequence of SEQ ID NO: 96, and/or a variable light chain comprising the amino acid sequence of SEQ ID NO: 97.

16 . The isolated antibody of claim 14 , wherein the heavy chain comprises an amino acid sequence having at least 99% sequence identity to SEQ ID NO: 98.

17 . The isolated antibody of claim 16 , wherein the heavy chain comprises the amino acid sequence of SEQ ID NO: 98.

18 . The isolated antibody of claim 14 , wherein the heavy chain comprises one to three constant region mutations.

19 . The isolated antibody of claim 18 , wherein the heavy chain comprises one constant region mutation.

20 . The isolated antibody of claim 14 , wherein the antibody comprises a light chain comprising the amino acid sequence of SEQ ID NO: 99.

21 . The isolated antibody of claim 18 , wherein the one to three constant region mutations enhance cell killing.

22 . The isolated antibody of claim 21 , wherein the enhanced cell killing is enhanced antibody-dependent cellular cytotoxicity (ADCC) and/or complement-dependent cytotoxicity (CDC).

23 . The isolated antibody of claim 18 , wherein the one to three constant region mutations are selected from L234, L235, G236, S239, F243, H268, D270, R292, S298, Y300, V305, K326, A330, 1332, E333, K334, and P396.

24 . The isolated antibody of claim 23 , wherein one to three constant region mutations are selected from S239D, S239M, F243L, H268D, D270E, R292P, S298A, Y300L, V305I, K326D, A330L, A330M, 1332E, E333A, K334A, K334E, and P396L.

25 . The isolated antibody of claim 23 , wherein the one to three constant region mutations are selected from: S239D/1332E, S239D/1332E/A330L, and S298A/E333A/K334A.

26 . The isolated antibody of claim 14 , wherein the antibody is afucosylated and/or galactosylated.

27 . An isolated antibody that binds human CCR8, wherein the antibody comprises:

a heavy chain complementarity determining region 1 (HCDR1) comprising the amino acid sequence of SEQ ID NO: 84, a heavy chain complementarity determining region 2 (HCDR2) comprising the amino acid sequence of SEQ ID NO: 85, a heavy chain complementarity determining region 3 (HCDR3) comprising the amino acid sequence of SEQ ID NO: 86, a light chain complementarity determining region 1 (LCDR1) comprising the amino acid sequence of SEQ ID NO: 87, a light chain complementarity determining region 2 (LCDR2) comprising the amino acid sequence of SEQ ID NO: 88, and a light chain complementarity determining region 3 (LCDR3) comprising the amino acid sequence of SEQ ID NO: 89;

wherein the antibody comprises a heavy chain comprising an amino acid sequence having at least 98% sequence identity to SEQ ID NO: 94, and a light chain comprising an amino acid sequence of SEQ ID NO: 95.

28 . The isolated antibody of claim 27 , wherein the heavy chain comprises one constant region mutation.

29 . An isolated antibody that binds human CCR8, wherein the antibody comprises:

a heavy chain complementarity determining region 1 (HCDR1) comprising the amino acid sequence of SEQ ID NO: 100, a heavy chain complementarity determining region 2 (HCDR2) comprising the amino acid sequence of SEQ ID NO: 85, a heavy chain complementarity determining region 3 (HCDR3) comprising the amino acid sequence of SEQ ID NO: 86, a light chain complementarity determining region 1 (LCDR1) comprising the amino acid sequence of SEQ ID NO: 87, a light chain complementarity determining region 2 (LCDR2) comprising the amino acid sequence of SEQ ID NO: 88, and a light chain complementarity determining region 3 (LCDR3) comprising the amino acid sequence of SEQ ID NO: 89;

wherein the antibody comprises a heavy chain comprising an amino acid sequence having at least 98% sequence identity to SEQ ID NO: 98, and a light chain comprising an amino acid sequence of SEQ ID NO: 99.

30 . The isolated antibody of claim 29 , wherein the heavy chain comprises one constant region mutation.
