Abstract

Compounds that inhibit p38α MAPK protein, and methods of using the same, are provided for treating or preventing diseases such as cancer or inflammatory diseases.

Claims (23)

1 . A method of treating an inflammatory disease in a patient comprising administering to a patient in need of such treatment a therapeutically effective amount of a compound having the formula:

wherein,

L 1 is selected from —CH 2 —, —C(CH 3 ) 2 —, and —C(CH 2 CH 2 )—; and

L 4 is selected from —NHCO—, —CONH—, —SO 2 NH—, —NHSO 2 —, and —CH═CH—.

2 . The method of claim 1 , wherein administering comprises orally administering.

The method of claim 1 , wherein administering comprises administering a dosage form comprising the compound, wherein the dosage form is selected from a controlled-release oral dosage form, a sustained-release oral dosage form, an extended-release oral dosage form, or a combination of any of the foregoing.

The method of claim 1 , wherein the inflammatory disease is selected from rheumatoid arthritis, a cardiovascular disease, multiple sclerosis, inflammatory bowel disease, inflammatory bowel diseases, chronic obstructive pulmonary disease, asthma, acute respiratory distress syndrome, and acute lung injury.

5 . The method of claim 1 , wherein the inflammatory disease is acute respiratory distress syndrome.

6 . A method of treating Alzheimer's disease, psoriasis, atherosclerosis, pain, multiple sclerosis, or glomerulosclerosis in a patient comprising administering to a patient in need of such treatment a therapeutically effective amount of a compound having the formula:

wherein,

L 1 is selected from —CH 2 —, —C(CH 3 ) 2 —, and —C(CH 2 CH 2 )—; and

L 4 is selected from —NHCO—, —CONH—, —SO 2 NH—, —NHSO 2 —, and —CH═CH—.

7 . The method of claim 6 , wherein administering comprises orally administering.

The method of claim 6 , wherein administering comprises administering a dosage form comprising the compound, wherein the dosage form is selected from a controlled-release oral dosage form, a sustained-release oral dosage form, an extended-release oral dosage form, or a combination of any of the foregoing.

9 . A method of treating cancer in a patient comprising administering to a patient in need of such treatment a therapeutically effective amount of a compound having the formula:

wherein,

L 1 is selected from —CH 2 —, —C(CH 3 ) 2 —, and —C(CH 2 CH 2 )—; and

L 4 is selected from —NHCO—, —CONH—, —SO 2 NH—, —NHSO 2 —, and —CH═CH—.

10 . The method of claim 9 , wherein administering comprises orally administering the pharmaceutical composition is for oral administration.

The method of claim 9 , wherein administering comprises administering a dosage form comprising the compound, wherein the dosage form is selected from a controlled-release oral dosage form, a sustained-release oral dosage form, an extended-release oral dosage form, or a combination of any of the foregoing.

12 . The method of claim 9 , wherein the cancer is associated with dysregulation of the p38α MAPK protein.

13 . The method of claim 9 , wherein the cancer is treatable by inhibiting the p38α MAPK protein.

14 . The method of claim 9 , wherein the cancer overexpresses the p38α MAPK protein.
