Abstract

Disclosed are formulations, including both liquid and lyophilized formulations, comprising a far-red dye probe and a non-linear surfactant or foamban. Also disclosed are related methods for preparing a lyophilized far-red dye probe formulation as well as related kits and diagnostic products.

Claims (19)

1 . A kit comprising:

(a) a first sealed container containing a lyophilized far-red dye probe formulation, wherein the lyophilized far-red dye probe formulation comprises

a far-red dye probe comprising a far-red dye conjugated to a carrier molecule, wherein the far-red dye is a far-red cyanine dye and the carrier molecule is an RNA;

a non-linear surfactant; and

at least one buffering agent;

and

(b) a second sealed container containing a diluent, wherein the diluent comprises the non-linear surfactant at a concentration of from 1% (v/v) to about 5% (v/v);

wherein the non-linear surfactant is present in the lyophilized far-red dye probe formulation and the diluent at concentrations configured to produce, upon reconstitution of the lyophilized formulation in the diluent, a final non-linear surfactant concentration of from about 1% (v/v) to about 10% (v/v).

2 . The kit of claim 1 , wherein the far-red cyanine dye is selected from the group consisting of cyanine 5 and cyanine 5.5.

3 . The kit of claim 1 , wherein the non-linear surfactant is selected from the group consisting of a polyoxyethylene sorbitan fatty acid ester and digitonin.

4 . The kit of claim 1 , wherein the non-linear surfactant is a polyoxyethylene sorbitan fatty acid ester selected from the group consisting of polysorbate 20, polysorbate 40, and polysorbate 60.

5 . The kit of claim 1 , wherein the non-linear surfactant is polysorbate 20.

6 . The kit of claim 1 , wherein the at least one buffering agent comprises Tris.

7 . The kit of claim 1 , wherein the far-red dye probe further comprises a quencher.

8 . The kit of claim 1 , wherein the far-red dye probe is selected from the group consisting of a molecular torch and a molecular beacon.

9 . The kit of claim 1 , wherein the lyophilized far-red dye probe formulation further comprises a promoter-based amplification oligomer.

10 . The kit of claim 5 , wherein the non-linear surfactant concentration in the diluent is 1% (v/v).

11 . The kit of claim 10 , wherein the non-linear surfactant is present in the lyophilized far-red dye probe formulation at a concentration configured to produce, upon reconstitution of the lyophilized formulation in the diluent, a final non-linear surfactant concentration of 1.41% (v/v) to about 2.24% (v/v).

12 . The kit of claim 11 , wherein the non-linear surfactant is present in the lyophilized far-red dye probe formulation at a concentration configured to produce, upon reconstitution of the lyophilized formulation in the diluent, a final non-linear surfactant concentration of 1.62% (v/v).
