Abstract

The present invention relates to immune cell-based anti-cancer therapeutics and methods of using the therapeutics in the treatment of cancer.

Claims (19)

1 . An isolated nucleic acid molecule encoding a polypeptide comprising from amino to carboxy terminus:

a tag-binding domain, wherein the tag-binding domain is an antibody or antigen-binding fragment thereof comprising a heavy chain variable domain comprising the amino acid sequence of SEQ ID NO: 33 and a light chain variable domain comprising the amino acid sequence of SEQ ID NO: 34, that binds to a tag comprising the amino acid sequence of SEQ ID NO: 44,

an extracellular hinge and transmembrane domain of CD28, and

a cytoplasmic signal transduction domain of CD28,

wherein the extracellular hinge and transmembrane domain of CD28 comprise amino acid sequences encoded by nucleotide sequences according to SEQ ID NOs: 6 and 7, respectively, and

wherein the cytoplasmic signal transduction domain of CD28 comprises an amino acid sequence encoded by a nucleotide sequence according to SEQ ID NO: 8.

2 . An isolated cell or an isolated vector comprising an isolated nucleic acid molecule according to claim 1 , wherein the cell is a T cell, a Natural Killer cell, or a cytotoxic T lymphocyte.

3 . A kit comprising:

the isolated vector according to claim 2 and a target module comprising a binding moiety, wherein the binding moiety is an antibody or an antigen-binding fragment thereof that specifically binds to CD2, CD3, CD4, CD8, CD10, CD19, CD20, CD22, CD23, CD33, CD38, CD44, CD52, CD99, CD123, CD274, TIM-3, erb1, erb2, erb3, erb4, EphA1-10, EphB1-6, PSCA, PSMA, carcinoembryonic antigen, fetal acetylcholine receptor, VEGFR1-3, epithelial cell adhesion molecule, alphafetoprotein, MUC1, MUC16, follicle stimulating hormone receptor, human high molecular weight-melanoma-associated antigen, folate binding protein, a-Folate receptor, ligands of the NKG2D receptor, EGP-2, EGP-4, CAIX, Ley, or wherein the binding moiety comprises alpha and beta chains or gamma and delta chains of a T cell receptor; and a tag comprising the amino acid sequence of SEQ ID NO: 44.

4 . The kit according to claim 3 , wherein the target module specifically binds to:

PSCA and comprises an amino acid sequence of SEQ ID NO: 28;

PSMA and comprises an amino acid sequence encoded by SEQ ID NO: 17;

CD33 and comprises an amino acid sequence of SEQ ID NO: 30;

CD123 and comprises an amino acid sequence of SEQ ID NO: 31; or

CD123×CD33 and comprises an amino acid sequence of SEQ ID NO: 32.

A formulation comprising the isolated cell according to claim 2 and a target module for administration to a subject, wherein said target module comprises a binding moiety, wherein the binding moiety is an antibody or an antigen-binding fragment thereof that specifically binds to CD2, CD3, CD4, CD8, CD10, CD19, CD20, CD22, CD23, CD33, CD38, CD44, CD52, CD99, CD123, CD274, TIM-3, erb1, erb2, erb3, erb4, EphA1-10, EphB1-6, PSCA, PSMA, carcinoembryonic antigen, fetal acetylcholine receptor, VEGFR1-3, epithelial cell adhesion molecule, alphafetoprotein, MUC1, MUC16, follicle stimulating hormone receptor, human high molecular weight-melanoma-associated antigen, folate binding protein, a-Folate receptor, ligands of the NKG2D receptor, EGP-2, EGP-4, CAIX, Ley, or wherein the binding moiety comprises alpha and beta chains or gamma and delta chains of a T cell receptor; and a tag comprising the amino acid sequence of SEQ ID NO: 44.

6 . The formulation according to claim 5 , wherein the antibody of the target module or antigen-binding fragment thereof specifically binds CD123.

7 . A method for stimulating an immune response in a mammal, comprising administering to the mammal an isolated cell according to claim 2 and a target module, wherein said target module comprises a binding moiety wherein the binding moiety is an antibody or an antigen-binding fragment thereof that specifically binds to CD2, CD3, CD4, CD8, CD10, CD19, CD20, CD22, CD23, CD33, CD38, CD44, CD52, CD99, CD123, CD274, TIM-3, erb1, erb2, erb3, erb4, EphA1-10, EphB1-6, PSCA, PSMA, carcinoembryonic antigen, fetal acetylcholine receptor, VEGFR1-3, epithelial cell adhesion molecule, alphafetoprotein, MUC1, MUC16, follicle stimulating hormone receptor, human high molecular weight-melanoma-associated antigen, folate binding protein, a-Folate receptor, ligands of the NKG2D receptor, EGP-2, EGP-4, CAIX, Ley, or wherein the binding moiety comprises alpha and beta chains or gamma and delta chains of a T cell receptor; and a tag comprising the amino acid sequence of SEQ ID NO: 44.

8 . The isolated nucleic acid molecule according to claim 1 , wherein the cytoplasmic signal transduction domain further comprises a CD3 zeta intracellular domain comprising an amino acid sequence encoded by a nucleotide sequence according to SEQ ID NO: 9.
