Abstract

Methods of suppressing the activation of microglial cells in the Central Nervous System (CNS), methods of ameliorating or treating the neurological effects of cerebral ischemia or cerebral inflammation, and methods of combating specific diseases that affect the CNS by administering a compound that binds to microglial receptors and prevents or reduces microglial activation are described. Also described are methods of screening compounds for the ability to suppress or reduce microglial activation.

Claims (21)

1. A method of suppressing microglial activation in the brain of a mammalian subject in need thereof, comprising administering to said subject a peptide of about 18 to 50 amino acids containing SEQ ID NO: 10 or a peptide consisting of SEQ ID NO: 10 in an amount effective to reduce microglial activation compared to that which would occur in the absence of the peptide, wherein microglial activation is suppressed.

2. The method of claim 1 , wherein said microglial activation is associated with CNS inflammation.

3. The method of claim 1 , wherein said microglial activation is associated with CNS ischemia.

4. The method of claim 1 , wherein said microglial activation is associated with closed head injury.

5. The method according to claim 2 , wherein said subject is afflicted with a condition selected from Alzheimer's disease and multiple sclerosis.

6. The method according to claim 3 , wherein said subject is afflicted with a condition selected from stroke, global cerebral ischemia, cerebral edema, closed acute head injury, and intracranial hemorrhage.

7. The method of claim 1 , wherein said peptide is linked to one to five additional amino acids or amino acid analogues at the N-terminus or C-terminus or both the N- and C-terminus, wherein such additional amino acids do not adversely affect the therapeutic function of the peptide.

8. The method of claim 1 , wherein said compound peptide contains about 18 amino acids.

9. The method of claim 1 , wherein said compound peptide contains about 20 amino acids.

10. The method of claim 1 , wherein said compound peptide contains about 30 amino acids.

11. The method of claim 1 , wherein said compound peptide contains about 40 amino acids.

12. The method of claim 1 , wherein said peptide consists essentially of SEQ ID NO. 10.

13. The method of claim 1 , further comprising measuring a neurologic outcome of the subject after administration of said peptide.

14. The method of claim 13 , wherein said neurologic outcome is measured by assessing a neurological status of said subject.

15. The method of claim 13 , wherein said neurologic outcome is measured by assessing one or more cellular effects.

16. The method of claim 15 , wherein said cellular effect is a histological effect.

17. The method of claim 15 , wherein said cellular effect is a morphometrical effect.

18. The method of claim 13 , wherein said neurologic outcome is a decrease in nitric oxide or nitrate levels.

19. The method of claim 13 , wherein said neurologic outcome is a decrease in inflammatory cytokines.

20. The method of claim 1 , wherein said peptide is conjugated to a carrier molecule that increases transport of said peptide across the blood-brain barrier, compared to that which would occur in the absence of said carrier molecule.

21. The method of claim 20 , wherein said carrier molecule is selected from the group consisting of pyridinium, fatty acids, inositol, cholesterol, glucose derivatives, hemoglobin, lysozyme, cytochrome c, ceruloplasmin, calmodulin, ubiquitin, substance P, histone, insulin and transferrin and peptides thereof, lipid vesicles and liposomes.
