Abstract

The present invention is directed to bendamustine esters and bendamustine amides and their use for the treatment of cancer.

Claims (30)

1. A pharmaceutical composition comprising a compound which is {5-[bis-(chloroethyl)-amino]-1-methyl-1H-benzimidazol-2-yl}butyric acid dodecyl ester having the chemical structure

or a pharmaceutically acceptable salt thereof, and a carrier, wherein said carrier comprises albumin.

2. The pharmaceutical composition according to claim 1 , wherein said compound, or a pharmaceutically acceptable salt thereof, and said albumin are formulated as nanoparticles.

3. The pharmaceutical composition according to claim 2 , wherein said nanoparticles have an average particle size of about 248 nm or less.

4. The pharmaceutical composition according to claim 1 , wherein the weight-to-weight ratio of said albumin to said compound, or a pharmaceutically acceptable salt thereof, is from about 2 to about 4.

5. The pharmaceutical composition according to claim 1 , wherein said albumin is human serum albumin.

6. The pharmaceutical composition according to claim 1 , wherein said pharmaceutical composition is in a solid form.

7. The pharmaceutical composition according to claim 1 , wherein said pharmaceutical composition is in a liquid form.

8. The pharmaceutical composition according to claim 1 , wherein said pharmaceutical composition is in a form suitable for intravenous administration to a patient in need thereof.

9. A pharmaceutical composition comprising a compound which is 4-{5-[bis-(chloroethyl)-amino]-1-methyl-1H-benzimidazol-2-yl}butyric acid dodecyl ester having the chemical structure

or a pharmaceutically acceptable salt thereof, and a carrier, wherein said carrier comprises albumin, prepared by a process comprising the steps of:

(a) preparing a first solution comprising a compound which is 4-{5-[bis-(chloroethyl)-amino]-1-methyl-1H-benzimidazol-2-yl}butyric acid dodecyl ester having the chemical structure,

or a pharmaceutically acceptable salt thereof, and at least one organic solvent;

(b) preparing a second solution comprising water and albumin;

(c) mixing said first solution and said second solution to afford an emulsion; and

(d) removing said organic solvent from said emulsion.

10. The pharmaceutical composition according to claim 9 , wherein said at least one organic solvent is dichloromethane.

11. The pharmaceutical composition according to claim 9 , wherein said albumin is human serum albumin.

12. The pharmaceutical composition according to claim 9 , wherein said organic solvent is dichloromethane and said albumin is human serum albumin.

13. A pharmaceutical composition, comprising

(a) a compound of which is 4-{5-[bis-(chloroethyl)-amino]-1-methyl-1H-benzimidazol-2-yl}butyric acid dodecyl ester having the chemical structure:

or a pharmaceutically acceptable salt thereof, and

(b) a means for increasing the circulation time of the compound in an aqueous environment.

14. A method of treating cancer in a patient, comprising administering to said patient a pharmaceutical composition comprising a compound which is 4-{5-[bis-(chloroethyl)-amino]-1-methyl-1H-benzimidazol-2-yl}butyric acid dodecyl ester having the chemical structure:

or a pharmaceutically acceptable salt thereof, and a carrier, wherein said carrier comprises albumin.

15. The method according to claim 14 , wherein said cancer is in the form of a solid tumor or a liquid tumor.

16. The method according to claim 15 , wherein said cancer is in the form of a solid tumor.

17. The method according to claim 16 , wherein said cancer is sarcoma, bladder cancer, cervical cancer, testicular cancer, melanoma, glioblastoma, colon cancer, head and neck cancer, ovarian cancer, prostate cancer, breast cancer, or lung cancer.

18. The method according to claim 15 , wherein said cancer is in the form of a liquid tumor.

19. The method according to claim 18 , wherein said cancer is lymphocytic leukemia, Hodgkin's disease, non-Hodgkin's lymphoma, multiple myeloma, or acute lymphocytic leukemia.
