Abstract

Described herein are neuroactive steroids or a pharmaceutically acceptable salt thereof. Such compounds are envisioned, in certain embodiments, to behave as GABA modulators. Also provided are pharmaceutical compositions comprising a compound described herein and methods of use and treatment, e.g., such as for inducing sedation and/or anesthesia.

Claims (30)

1 . A compound of Formula (1-A):

or a pharmaceutically acceptable salt thereof, wherein:

R 3 is alkyl, alkenyl, carbocyclyl, heterocyclyl, aryl, or heteroaryl;

one of R X and R Y is aryl and the other is hydrogen or alkyl;

R 19 is hydrogen or alkyl;

R 5 is hydrogen;

represents a single bond; and

a is 1.

2 . The compound of claim 1 , wherein R 3 is alkyl.

3 . The compound of claim 2 , wherein the alkyl is a substituted alkyl.

4 . The compound of claim 1 , wherein one of R X and R Y is aryl and the other is hydrogen.

5 . The compound of claim 1 , wherein one of R X and R Y is aryl and the other is alkyl.

6 . The compound of claim 1 , wherein R 19 is hydrogen.

7 . The compound of claim 1 , wherein R 19 is alkyl.

8 . The compound of claim 7 , wherein the alkyl is a substituted alkyl.

9 . A compound of the Formula (III-b):

or a pharmaceutically acceptable salt thereof, wherein:

R 3 is alkyl, alkenyl, carbocyclyl, heterocyclyl, aryl, or heteroaryl;

each of R 2 , R 4 , R 6 , R 7 , R 11a , and R 11b is independently hydrogen;

Q is —N(R A1 )(R A1 );

each instance of R A1 is independently hydrogen, alkyl, alkenyl, carbocyclyl, heterocyclyl, aryl, or heteroaryl;

R 19 is —C(R C ) 2 OR A1 , wherein R C is hydrogen or alkyl; and

represents a single bond and R 5 is hydrogen.

10 . A compound, or pharmaceutically acceptable salt thereof, selected from the group consisting of:

11 . A pharmaceutical composition comprising a compound of claim 1 and a pharmaceutically acceptable excipient.

12 . A method for reducing the severity of autism spectrum disorder in a subject in need thereof, comprising administering to the subject a therapeutically effective amount of a compound of claim 1 .

13 . A kit comprising a solid composition comprising a compound of claim 1 and a sterile diluent.

14 . A pharmaceutical composition comprising a compound of claim 10 and a pharmaceutically acceptable excipient.

15 . A method for reducing the severity of autism spectrum disorder in a subject in need thereof, comprising administering to the subject a therapeutically effective amount of a compound of claim 10 .

16 . A kit comprising a solid composition comprising a compound of claim 10 and a sterile diluent.
