Abstract

The present invention relates to a novel human gene encoding a polypeptide which is a member of the TNF receptor family, and has now been found to bind TRAIL. More specifically, an isolated nucleic acid molecule is provided encoding a human polypeptide named tumor necrosis factor receptor-5, sometimes referred to as “TNFR-5” or “TR5,” and now referred to hereinafter as “TRAIL receptor without intracellular domain” or “TRID.” TRID polypeptides are also provided, as are vectors, host cells, and recombinant methods for producing the same. The invention further relates to screening methods for identifying agonists or antagonists of TRAIL polypeptide activity. Also provided are diagnostic and therapeutic methods utilizing such compositions.

Claims (23)

1. An isolated polypeptide comprising amino acids 1 to 214 of SEQ ID NO:2.

2. The polypeptide of claim 1 , consisting of amino acids 1 to 214 of SEQ ID NO:2.

3. The polypeptide of claim 1 , which is produced by a recombinant host cell.

4. The polypeptide of claim 3 , wherein said recombinant host cell is a eukaryotic host cell.

5. The polypeptide of claim 1 , wherein said polypeptide is fused to a heterologous polypeptide.

6. The polypeptide of claim 5 , wherein said heterologous polypeptide comprises an immunoglobulin Fc region.

7. The polypeptide of claim 6 , wherein said Fc region is a human immunoglobulin Fc region.

8. A composition comprising the polypeptide of claim 1 , and a carrier.

9. An isolated polypeptide comprising amino acids 27 to 123 of SEQ ID NO:2, wherein said polypeptide inhibits apoptosis.

10. The polypeptide of claim 9 , which is produced by a recombinant host cell.

11. The polypeptide of claim 10 , wherein said recombinant host cell is a eukaryotic host cell.

12. The polypeptide of claim 9 , wherein said polypeptide is fused to a heterologous polypeptide.

13. The polypeptide of claim 12 , wherein said heterologous polypeptide comprises an immunoglobulin Fc region.

14. The polypeptide of claim 13 , wherein said Fc region is a human immunoglobulin Fc region.

15. A composition comprising the polypeptide of claim 9 , and a carrier.

16. An isolated polypeptide comprising an amino acid sequence that is at least 90% identical to amino acids 1 to 214 of SEQ ID NO:2, wherein said polypeptide inhibits apoptosis.

17. The polypeptide of claim 16 comprising an amino acid sequence that is at least 95% identical to amino acids 1 to 214 of SEQ ID NO:2.

18. The polypeptide of claim 16 , which is produced by a recombinant host cell.

19. The polypeptide of claim 18 , wherein said recombinant host cell is a eukaryotic host cell.

20. The polypeptide of claim 16 , wherein said polypeptide is fused to a heterologous polypeptide.

21. The polypeptide of claim 20 , wherein said heterologous polypeptide comprises an immunoglobulin Fc region.

22. The polypeptide of claim 21 , wherein said Fc region is a human immunoglobulin Fc region.

23. A composition comprising the polypeptide of claim 16 , and a carrier.
