Abstract

The present invention relates to intracellular signaling molecules, in particular the Nod2 protein and nucleic acids encoding the Nod2 protein. The present invention provides methods of identifying modulators of Nod2 signaling. In particular, the present invention additionally provides methods of screening immune modulators such as adjuvants using Nod2. The present invention further provides methods of altering Nod2 signaling.

Claims (29)

1. A method of screening compounds, comprising:

a) providing

i) a cell expressing a Nod2 variant, wherein said Nod2 variant is selected from the group consisting of SEQ ID NOs: 55, 57, 59, and 61, and wherein said Nod2 variant is defective in responding to bacterial muropeptides; and

ii) a plurality of test compounds; and

b) contacting said cell with said plurality of test compounds; and

c) determining the presence or absence of a response to a bacterial muropeptide.

2. The method of claim 1 , wherein said response to a bacterial muropeptide comprises the activation of NF-κB.

3. The method of claim 2 , wherein said activation of NF-κB is detected using a reporter gene assay.

4. The method of claim 1 , wherein said Nod2 variant is a Crohn's disease associated variant.

5. The method of claim 1 , wherein said cell expressing a Nod2 variant comprises peripheral blood mononuclear cells isolated from an individual diagnosed with Crohn's disease.

6. The method of claim 5 , wherein said response to a bacterial muropeptide comprises the activation of NF-κB.

7. The method of claim 6 , wherein said activation of NF-κB is detected using an electrophoretic mobility shift assay.

8. The method of claim 1 , wherein said test compound is a drug.

9. The method of claim 1 , wherein said test compound is a muramyl dipeptide.

10. The method of claim 9 , wherein said muramyl dipeptide is selected from the group consisting of MurNAc-L-Ala-D-isoGln, MuraNac-L-Ala-D-isoGln, MuraNac-L-Ala-D-Glu, analogs of MurNAc-L-Ala-D-isoGln, analogs of MuraNac-L-Ala-D-isoGln, and analogs of MuraNac-L-Ala-D-Glu.

11. A method of screening compounds, comprising

a) providing

i) a cell expressing Nod2; and

ii) a plurality of test compounds; and

b) contacting said cell with said plurality of test compounds; and

c) determining the level of Nod2 activity in said cell in response to said test compound.

12. The method of claim 11 , wherein said Nod2 activity comprises activation of NF-κB.

13. The method of claim 12 , wherein said activation of NF-κB is detected using a reporter gene assay.

14. The method of claim 11 , wherein said test compound is a muramyl dipeptide.

15. The method of claim 14 , wherein said muramyl dipeptide is selected from the group consisting of MurNAc-L-Ala-D-isoGln, MuraNac-L-Ala-D-isoGln, MuraNac-L-Ala-D-Glu, analogs of MurNAc-L-Ala-D-isoGln, analogs of MuraNac-L-Ala-D-isoGln, and analogs of MuraNac-L-Ala-D-Glu.

16. The method of claim 11 , wherein said test compound is a vaccine adjuvant.

17. The method of claim 11 , wherein said test compound in an anti-adjuvant.

18. The method of claim 11 , wherein said Nod2 activity is increased in response to said test compound.

19. The method of claim 11 , wherein said Nod2 activity is decreased in response to said test compound.
