Abstract

Methods and apparatus for direct detection of chemical reactions are provided. Electric charge perturbations of the local environment during enzyme-catalyzed reactions are sensed by an electrode system with an immobilized target molecule. The charge perturbation caused by the polymerase reaction can uniquely identify a DNA sequence. The polymerization process generates local perturbations of charge in the solution near the electrode surface and induces a charge in a polarazible gold electrode. This event is detected as a transient current by a voltage clamp amplifier. Detection of single nucleotides in a sequence can be determined by dispensing individual dNTPs to the electrode solution and detecting the charge perturbations. Alternatively, multiple bases can be determined at the same time using a mix of all dNTPs with subsequent analysis of the resulting signal. This technique may be adapted to other reaction determinations, such as enzymatic reactions, other electrode configurations, and other amplifying circuits.

Claims (23)

1. A sensor comprising:

a polarizable electrode that is polarizable due to a charge perturbations in a reaction medium;

an insulating layer on a surface of the polarizable electrode;

at least one template nucleic acid linked to the insulating layer;

and

circuitry to apply a first voltage to the polarizable electrode, and to detect a current pulse through the polarizable electrode in response to incorporation of nucleotides into the at least one template nucleic acid, said circuitry comprising differential feedback amplifier having one input at a fixed voltage, said input located outside a container containing the reaction medium, and another input attached to the polarizable electrode.

2. The sensor of claim 1 , wherein the sensor is in a reaction medium containing nucleotides in a solution contacting the insulating layer.

3. The sensor of claim 1 , wherein the circuitry includes a feedback loop to apply the first voltage to the electrode.

4. The sensor of claim 1 , wherein the circuitry is connected to the electrode and thereby coupled to the template nucleic acid to detect a current pulse induced in response to charged particles liberated during the incorporation.

5. The sensor of claim 1 , wherein the circuitry is connected to the electrode and thence to the template nucleic acid to detect a current pulse induced in response to a change in surface charge of the insulating layer due to the incorporation.

6. The sensor of claim 1 , wherein the sensor is in a reaction medium containing nucleotides that are label-free.

7. The sensor of claim 1 , wherein the circuitry measures a plurality of current pulses and correlates them with the nucleotides to indicate at least a portion of a sequence of at least a portion of the template nucleic acid.

8. The sensor of claim 1 , further comprising a microfluidic structure to provide the nucleotides.

9. The sensor of claim 1 , further comprising a second electrode to provide a reference signal, and wherein the circuitry compares the detected current pulse to the reference signal to detect occurrence of the incorporation.

10. A sensor comprising:

a polarizable electrode that is polarizable due to charge perturbations in a reaction medium;

an insulating layer on a surface of the electrode;

at least one template nucleic acid linked to the insulating layer;

a circuit to apply a first voltage to the polarizable electrode and comprising a differential feedback amplifier having one input at a fixed voltage outside a container for containing the reaction medium and another input attached to the polarizable electrode; and

a detector to detect a current pulse through the polarizable electrode in response to incorporation of nucleotides into the at least one template nucleic acid.

11. The sensor of claim 10 , wherein the sensor is in a reaction medium having nucleotides contacting the insulating layer.

12. The sensor of claim 10 , wherein the circuitry includes a feedback loop to apply the first voltage to the electrode.

13. The sensor of claim 10 , wherein the circuitry detects a current pulse induced in response to charged particles liberated during the incorporation.
