Abstract

Hydrogels comprising a macromolecular matrix and water may be used for aesthetic fillers, for example, dermal fillers. The macromolecular matrix may include a crosslinked combination of hyaluronic acid and collagen.

Claims (57)

1. A method of improving an aesthetic quality of a soft tissue of a human subject, the method comprising:

(a) providing adipose tissue;

(b) providing a hydrogel composition comprising:

(i) water; and

(ii) a hyaluronic acid crosslinked with a collagen via a plurality of crosslink units,

wherein at least a portion of the crosslink units comprise an amide bond; and

wherein the hyaluronic acid concentration is from 6 mg/mL to 20 mg/mL;

the collagen concentration is from 3 mg/mL to 12 mg/mL;

the hyaluronic acid:collagen weight ratio is from 1:1 to 7:1; and

the hydrogel has a storage modulus value of between 850 Pa and 5,000 Pa;

(c) contacting the hydrogel composition with the adipose tissue ex vivo to provide a mixture; and

(d) administering the mixture to the soft tissue of the subject;

thereby improving the aesthetic quality of the soft tissue.

2. The method of claim 1 , wherein the collagen is collagen type I, collagen type III, or a combination thereof.

3. The method of claim 1 , wherein the collagen is collagen type I.

4. The method of claim 2 or 3 , wherein the collagen is derived from a human or porcine source.

5. The method of claim 1 , wherein the hyaluronic acid has a molecular weight of about 1,000,000 Da to about 5,000,000 Da.

6. The method of claim 1 , wherein the hyaluronic acid has a molecular weight of about 1,000,000 Da to about 3,000,000 Da.

7. The method of claim 1 , wherein the ratio of the hyaluronic acid to the collagen is 1:1 to 2:1.

8. The method of claim 1 , wherein the ratio of the hyaluronic acid to the collagen is 1:1.

9. The method of claim 1 , wherein the hydrogel has a storage modulus value of between 1,000 Pa and 5,000 Pa.

10. The method of claim 1 , wherein the hydrogel has a hyaluronic acid concentration of 12 mg/mL or 16 mg/mL.

11. The method of claim 1 , wherein the hydrogel has a collagen concentration of 6 mg/mL, 8 mg/mL or 12 mg/mL.

12. The method of claim 1 , wherein the hydrogel has a hyaluronic acid concentration of 12 mg/mL and a collagen concentration of 6 mg/mL.

13. The method of claim 1 , wherein the hydrogel has a hyaluronic acid concentration of 12 mg/mL and a collagen concentration of 12 mg/mL.

14. The method of claim 1 , wherein the hydrogel has a hyaluronic acid concentration of 16 mg/mL and a collagen concentration of 8 mg/mL.

15. The method of claim 1 , wherein the administration is intradermal.

16. The method of claim 1 , wherein the administration is subcutaneous.

17. The method of claim 1 , wherein the administration is by injection.

18. A method of improving an aesthetic quality of a soft tissue of a human subject, the method comprising:

(a) administering a hydrogel composition to the soft tissue of the subject, wherein the hydrogel composition comprises:

(i) water; and

(ii) a hyaluronic acid crosslinked with a collagen via a plurality of crosslink units,

wherein at least a portion of the crosslink units comprise an amide bond; and

wherein the hyaluronic acid concentration is from 6 mg/mL to 20 mg/mL;

the collagen concentration is from 3 mg/mL to 12 mg/mL;

the hyaluronic acid:collagen weight ratio is from 1:1 to 7:1; and

the hydrogel has a storage modulus value of between 850 Pa and 5,000 Pa;

and

(b) contacting the hydrogel composition with adipose tissue in vivo;

thereby improving the aesthetic quality of the soft tissue.

19. The method of claim 18 , wherein the collagen is collagen type I, collagen type III, or a combination thereof.

20. The method of claim 18 , wherein the collagen is collagen type I.

21. The method of claim 19 or 20 , wherein the collagen is derived from a human or porcine source.

22. The method of claim 18 , wherein the hyaluronic acid has a molecular weight of about 1,000,000 Da to about 5,000,000 Da.

23. The method of claim 18 , wherein the hyaluronic acid has a molecular weight of about 1,000,000 Da to about 3,000,000 Da.

24. The method of claim 18 , wherein the ratio of the hyaluronic acid to the collagen is 1:1 to 2:1.

25. The method of claim 18 , wherein the ratio of the hyaluronic acid to the collagen is 1:1.

26. The method of claim 18 , wherein the hydrogel has a storage modulus value of between 1,000 Pa and 5,000 Pa.

27. The method of claim 18 , wherein the hydrogel has a hyaluronic acid concentration of 12 mg/mL or 16 mg/mL.

28. The method of claim 18 , wherein the hydrogel has a collagen concentration of 6 mg/mL, 8 mg/mL or 12 mg/mL.

29. The method of claim 18 , wherein the hydrogel has a hyaluronic acid concentration of 12 mg/mL and a collagen concentration of 6 mg/mL.

30. The method of claim 18 , wherein the hydrogel has a hyaluronic acid concentration of 12 mg/mL and a collagen concentration of 12 mg/mL.

31. The method of claim 18 , wherein the hydrogel has a hyaluronic acid concentration of 16 mg/mL and a collagen concentration of 8 mg/mL.

32. The method of claim 18 , wherein the administration is intradermal.

33. The method of claim 18 , wherein the administration is subcutaneous.

34. The method of claim 18 , wherein the administration is by injection.
