Abstract

The invention relates to inhibitory nucleic acids and rAAV-based compositions, methods and kits useful for treating Amyotrophic Lateral Sclerosis.

Claims (14)

1. A synthetic miRNA comprising 5, 6, 7, 8, 9, 10, 11, 12, 13, 14, 15, 16, 17, 18, 19, 20, or 21 consecutive nucleotides encoded by a sequence set forth in SEQ ID NO: 17, and flanking regions of miR-155.

2. A composition comprising the synthetic miRNA of claim 1 .

3. A recombinant nucleic acid comprising a sequence set forth in SEQ ID NO: 17, flanking regions of miR-155, and further comprising an inverted terminal repeat (ITR) of an Adeno-Associated Virus (AAV) serotype.

4. The recombinant nucleic acid of claim 3 , wherein the AAV serotype is selected from the group consisting of AAV1, AAV2, AAV5, AAV6, AAV6.2, AAV7, AAV8, AAV9, AAVRh10, AAV11, and variants thereof.

5. The recombinant nucleic acid of claim 4 , further comprising a promoter operably linked with a region(s) encoding the miRNA.

6. The recombinant nucleic acid of claim 5 , wherein the promoter is a tissue-specific promoter.

7. The recombinant nucleic acid of claim 6 , wherein the promoter is a polymerase II promoter, such as a β-actin promoter.

8. The recombinant nucleic acid of claim 6 , wherein the promoter is a polymerase III promoter, such as a U6 promoter.

9. A composition comprising the recombinant nucleic acid of claim 3 .

10. A recombinant Adeno-Associated Virus (AAV) comprising a recombinant nucleic acid encoding a miRNA comprising 5, 6, 7, 8, 9, 10, 11, 12, 13, 14, 15, 16, 17, 18, 19, 20, or 21 consecutive nucleotides encoded by a sequence as set forth in SEQ ID NO: 17, flanking regions of miR-155, and further comprising an inverted terminal repeat (ITR) of an AAV serotype.

11. The recombinant AAV of claim 10 , further comprising one or more capsid proteins of one or more AAV serotypes selected from the group consisting of: AAV1, AAV2, AAV3, AAV4, AAV5, AAV6, AAV7, AAV8, AAV9, AAV.Rh10, AAV11 2 and variants thereof.

12. A composition comprising the recombinant AAV of claim 10 .

13. The composition of claim 12 , further comprising a pharmaceutically acceptable carrier.

14. A kit comprising a container comprising the composition of claim 12 .
