Abstract

The present invention provides binding polypeptides (e.g., antibodies or fragments thereof) that specifically bind to a target antigen (e.g., a human antigen, e.g., human PDGFRβ) with high affinity. The invention also provides, libraries of binding polypeptides, pharmaceutical compositions, as well as nucleic acids encoding binding polypeptides, recombinant expression vectors and host cells for making such binding polypeptides. Methods of using binding polypeptide of the invention to diagnose and treat disease are also encompassed by the invention.

Claims (28)

1. An isolated nucleic acid encoding a binding polypeptide that specifically binds to PDGFRβ, wherein the binding polypeptide comprises a VH domain and a VL domain, wherein

(i) the VH domain comprises a CDR3 comprising the amino acid sequence set forth in SEQ ID NO: 1, a CDR2 comprising the amino acid sequence set forth in SEQ ID NO: 3, and a CDR1 comprising the amino acid sequence set forth in SEQ ID NO: 35; and

(ii) the VL domain comprises a CDR3 comprising the amino acid sequence set forth in SEQ ID NO: 119, a CDR2 comprising the amino acid sequence set forth in SEQ ID NO: 204, and a CDR1 comprising the amino acid sequence set forth in SEQ ID NO: 289.

2. A recombinant expression vector comprising the nucleic acid of claim 1 .

3. An isolated host cell comprising the recombinant expression vector of claim 2 .

4. A method of producing a binding polypeptide that binds specifically to human PDGFRβ, comprising culturing the host cell of claim 3 under conditions such that a binding polypeptide that binds specifically to human PDGFRβ is produced by the host cell.

5. The isolated nucleic acid of claim 1 , wherein the binding polypeptide comprises a VH domain comprising an amino acid sequence sharing at least 95% sequence identity with SEQ ID NO: 321 and a VL domain comprising an amino acid sequence sharing at least 95% sequence identity with SEQ ID NO: 425.

6. The isolated nucleic acid of claim 1 , wherein the binding polypeptide comprises a VH domain comprising the amino acid sequence set forth in SEQ ID NO: 321 and a VL domain comprising the amino acid sequence set forth in SEQ ID NO: 425.

7. The isolated nucleic acid of claim 1 , wherein the binding polypeptide inhibits activity of PDGFRβ.

8. The isolated nucleic acid of claim 1 , wherein the binding polypeptide binds to PDGFRβ with a Kd of less than 250 pM.

9. The isolated nucleic acid of claim 1 , wherein the binding polypeptide binds to PDGFRβ with a Kd of about 100 pM.

10. The isolated nucleic acid of claim 1 , wherein the binding polypeptide binds to PDGFRβ with an off-rate of about 10 −3 s −1 as measured by surface plasmon resonance (SPR).

11. The isolated nucleic acid of claim 1 , wherein the binding polypeptide binds specifically to mouse and human PDGFRβ.

12. The isolated nucleic acid of claim 1 , wherein the binding polypeptide is an antibody.

13. The isolated nucleic acid of claim 1 , wherein the binding polypeptide is an scFv.

14. An isolated nucleic acid encoding a binding polypeptide that specifically binds to PDGFRβ, wherein the binding polypeptide comprises a VH domain and a VL domain, wherein

(i) the VH domain comprises a CDR3 comprising the amino acid sequence set forth in SEQ ID NO: 1, a CDR2 comprising the amino acid sequence set forth in SEQ ID NO: 2, and a CDR1 comprising the amino acid sequence set forth in SEQ ID NO: 33; and

(ii) the VL domain comprises a CDR3 comprising the amino acid sequence set forth in SEQ ID NO: 98, a CDR2 comprising the amino acid sequence set forth in SEQ ID NO: 183, and a CDR1 comprising the amino acid sequence set forth in SEQ ID NO: 268.

15. A recombinant expression vector comprising the nucleic acid of claim 14 .

16. An isolated host cell comprising the recombinant expression vector of claim 15 .

17. A method of producing a binding polypeptide that binds specifically to human PDGFRβ, comprising culturing the host cell of claim 16 under conditions such that a binding polypeptide that binds specifically to human PDGFRβ is produced by the host cell.

The isolated nucleic acid of claim 14 , wherein the binding polypeptide comprises a VH domain comprising an amino acid sequence sharing at least 95% sequence identity with SEQ ID NO: 318 and a VL domain comprising an amino acid sequence sharing at least 95% sequence identity with SEQ ID NO: 425.

19. The isolated nucleic acid of claim 14 , wherein the binding polypeptide comprises a VH domain comprising the amino acid sequence set forth in SEQ ID NO: 318 and a VL domain comprising the amino acid sequence set forth in SEQ ID NO: 425.

20. The isolated nucleic acid of claim 14 , wherein the binding polypeptide inhibits activity of PDGFRβ.

21. The isolated nucleic acid of claim 14 , wherein the binding polypeptide binds to PDGFRβ with a Kd of less than 15 nM.

22. The isolated nucleic acid of claim 14 , wherein the binding polypeptide binds specifically to mouse and human PDGFRβ.

23. The isolated nucleic acid of claim 14 , wherein the binding polypeptide is an antibody.

24. The isolated nucleic acid of claim 14 , wherein the binding polypeptide is an scFv.
