Abstract

The present invention relates generally to systems and methods for measuring an analyte in a host. More particularly, the present invention relates to systems and methods for transcutaneous and subcutaneous measurement of glucose in a host.

Claims (35)

1. An analyte sensor system comprising:

a transcutaneous analyte sensor body comprising an electrode configured to continuously measure a glucose level in a host for a plurality of days;

a membrane disposed over the electrode, the membrane comprising:

a first layer configured to limit transport of glucose to the electrode, the first layer including a copolymer comprising polyurethane and silicone; and

a second layer comprising an enzyme to catalyze a reaction of glucose and oxygen as a co-reactant; and

a sensor electronics unit operably connected to the electrode and configured to measure a current produced by the electrode during the plurality of days, wherein the system is configured to measure a signal indicative of a glucose concentration with substantial linearity at glucose concentrations of up to about 400 mg/dL at an oxygen concentration of 0.6 mg/L.

2. The analyte sensor system of claim 1 , wherein the sensor electronics unit is configured to directly measure the current produced by the electrode.

3. The analyte sensor system of claim 1 , further comprising a biointerface membrane configured to support tissue ingrowth.

4. The analyte sensor system of claim 1 , further comprising an analog-to-digital converter configured to translate the current into a digital signal.

5. The analyte sensor system of claim 1 , wherein the electrode comprises an exposed electroactive working electrode surface with a surface area of from about 0.00002 in 2 to about 0.0079 in 2 .

6. The analyte sensor system of claim 1 , wherein the membrane comprises a resistance domain configured to have a permeability ratio of at least about 50:1 of glucose to an interferant.

7. The analyte sensor system of claim 1 , wherein the membrane comprises a resistance domain configured to have a permeability ratio of at least about 200:1 of glucose to an interferant.

8. The analyte sensor system of claim 1 , wherein the sensor system is configured to have, in operation, a sensitivity of from about 5 pA/mg/dL to about 25 pA/mg/dL.

9. The analyte sensor system of claim 1 , wherein the sensor system is configured to have, in operation, a sensitivity of from about 3.5 to about 7.5 pA/mg/dL.

10. The analyte sensor system of claim 1 , wherein the sensor system is configured to have, in operation, a sensitivity of from about 1 pA/mg/dL to about 100 pA/mg/dL.

11. The analyte sensor system of claim 1 , wherein the first layer is formed from a polyurethane membrane comprising both hydrophilic and hydrophobic regions.

12. The analyte sensor system of claim 1 , further comprising the electrode being configured to continuously measure a glucose level at a plurality of intervals during the plurality of days.

13. The analyte sensor system of claim 1 , wherein the first layer comprises polyurethaneurea.

14. The analyte sensor system of claim 1 , wherein the first layer and/or second layer comprises polyvinylpyrrolidone.

15. An analyte sensor system comprising:

a transcutaneous analyte sensor body comprising an electrode configured to continuously measure a glucose level in a host for a plurality of days;

a membrane disposed over the electrode, the membrane comprising:

a first layer configured to limit transport of glucose to the electrode, the first layer including a copolymer comprising polyurethane and silicone; and

a second layer comprising an enzyme to catalyze a reaction of glucose and oxygen as a co-reactant, the second layer including polyvinylpyrrolidone; and

a sensor electronics unit operably connected to the electrode and configured to measure a current produced by the electrode during the plurality of days, wherein the system is configured to measure a signal indicative of a glucose concentration with substantial linearity at glucose concentrations of up to about 400 mg/dL.

16. The sensor system of claim 15 , wherein the electrode comprises a surface area from about 0.000084 cm{circumflex over ( )}2 to about 0.016 cm{circumflex over ( )}2.

17. The sensor system of claim 15 , further comprising the first layer comprising polyurethaneurea.

18. An analyte sensor system comprising:

a transcutaneous analyte sensor body comprising an electrode configured to continuously measure a glucose level in a host for a plurality of days;

a membrane disposed over the electrode, the membrane comprising:

a first layer configured to limit transport of glucose to the electrode, the first layer including a copolymer comprising polyurethane and silicone, and a hydrophilic component, wherein the hydrophilic component is polyvinylpyrrolidone; and

a second layer comprising an enzyme to catalyze a reaction of glucose and oxygen as a co-reactant; and

a sensor electronics unit operably connected to the electrode and configured to measure a current produced by the electrode during the plurality of days, wherein the system is configured to measure a signal indicative of a glucose concentration with substantial linearity at glucose concentrations of up to about 400 mg/dL.

19. The sensor system of claim 18 , wherein the electrode comprises a surface area from about 0.000084 cm{circumflex over ( )}2 to about 0.016 cm{circumflex over ( )}2.

20. The sensor system of claim 18 , wherein the system is configured to measure the signal indicative of the glucose concentration with substantial linearity at an oxygen concentration of 0.6 mg/L.
