Abstract

Ultrasound transducer element arrays using acoustic ensonification drive patterns via a patient interface for sonosensitizer activation in sonodynamic therapy. Incoherent acoustic field generation varying phase, frequencies, and/or amplitude via controlled delivery of low intensity planar acoustic waves. Method includes generating a first and a second signal to generate respective acoustic ensonification drive patterns with phase, frequency, and amplitude and generating at least one relative phase, frequency or amplitude difference to generate a third incoherent acoustic ensonification pattern to activate a sonosensitizer. Calibration and complementary therapy procedures to improve cell susceptibility of cells to sonodynamic therapy are disclosed.

Claims (44)

1. An ultrasound transducer system for applying sonodynamic therapy to an anatomical structure, the ultrasound transducer system comprising:

an ultrasonic transducer array of piezoelectric transducers, wherein each piezoelectric transducer in the ultrasonic transducer array comprises an emitting surface configured to emit planar or defocused acoustic waves,

wherein the ultrasonic transducer array is configured for calibration based on at least one of transcranial transmission data, imaging data, and acoustic pulses, wherein the acoustic pulses are generated by one or more of a chirp signal, an impulse signal, and a ping signal, wherein the ping signal comprises a burst of a plurality of acoustic pulses at a calibration frequency;

a patient interface comprising a cooling system, the patient interface being configured to acoustically couple the ultrasonic transducer array to a patient,

wherein the patient interface is configured to detachably couple to the ultrasonic transducer array,

wherein the ultrasonic transducer array projects to the patient interface through a cavity, wherein the cavity is configured to be filled with a fluid to a predetermined volume,

wherein the patient interface comprises an acoustically-neutral elastic membrane,

wherein the acoustically-neutral elastic membrane is configured to inflate in order to couple to at least two alignment features configured to receive predetermined anatomical landmarks in order to establish a location registration between at least two of the ultrasonic transducer array, the acoustically-neutral elastic membrane, and the anatomical structure,

wherein the cooling system comprises one or more cooling channels configured for receiving the fluid,

wherein the one or more cooling channels are configured for circulation of the fluid to reduce heat at the acoustically-neutral elastic membrane; and

a controller coupled to the ultrasonic transducer array,

wherein the controller is configured to drive the ultrasonic transducer array with one or more frequencies selected from a range of 20 kHz to 2 MHZ,

wherein the controller is configured to drive the ultrasonic transducer array with a signal configured to minimize a spatial variation of an acoustic wave intensity to cause the ultrasonic transducer array to activate a sonosensitizer in a treatment region in the anatomical structure with the planar or defocused acoustic waves,

wherein the signal is configured with one or more duty cycles to drive each piezoelectric transducer in the ultrasonic transducer array to generate a high temporal peak acoustic intensity with a low temporal average acoustic intensity when activating the sonosensitizer at the treatment region in the anatomical structure with the planar or defocused acoustic waves.

2. The ultrasound transducer system of claim 1 , wherein the anatomical structure is a patient's head, and wherein the at least two alignment features are configured to receive predetermined anatomical landmarks of the patient's head.

3. The ultrasound transducer system of claim 1 , wherein the predetermined anatomical landmarks comprise a zygomatic arch, a mastoid tip, middle arch of eyebrows, or combinations thereof.

4. The ultrasound transducer system of claim 1 , wherein the patient interface comprises a receptacle configured to releasably couple to the ultrasonic transducer array.

The ultrasound transducer system of claim 1 , wherein the patient interface comprises spaced apart receptacles, and wherein the ultrasonic transducer array is selectively configured to releasably couple to the spaced apart receptacles to achieve an optimal treatment position for the ultrasonic transducer array.

6. The ultrasound transducer system of claim 1 , wherein the patient interface is configured to acoustically couple the ultrasonic transducer array to the patient, wherein the acoustically-neutral elastic membrane is configured to conform to the anatomical structure.

7. The ultrasound transducer system of claim 1 , wherein the acoustically-neutral elastic membrane is positioned over an exit plane of the ultrasonic transducer array.

8. The ultrasound transducer system of claim 1 , wherein the fluid is degassed.

9. The ultrasound transducer system of claim 1 , wherein the fluid is chilled to remove residual heat during treatment.

10. An ultrasound transducer system for applying sonodynamic therapy to an anatomical structure, the ultrasound transducer system comprising:

an ultrasonic transducer array of piezoelectric transducers,

wherein the ultrasonic transducer array is configured for calibration based on at least one of transcranial transmission data, imaging data, and acoustic pulses, wherein the acoustic pulses are generated by one or more of a chirp signal, an impulse signal, and a ping signal, wherein the ping signal comprises a burst of a plurality of acoustic pulses at a calibration frequency,

an array holder,

a patient interface,

a cooling system, and

a controller;

wherein each piezoelectric transducer in the ultrasonic transducer array comprises an emitting surface configured to emit planar or defocused acoustic waves;

wherein the array holder is configured to couple to the ultrasonic transducer array, wherein the array holder is configured to adjust to move the ultrasonic transducer array relative to the patient interface between a plurality of treatment positions;

wherein the ultrasonic transducer array projects toward the patient interface through a cavity, wherein the cavity is configured to be filled with a fluid to a predetermined volume,

wherein the patient interface is configured to acoustically couple the ultrasonic transducer array to a patient,

wherein the patient interface is configured to detachably couple to the ultrasonic transducer array,

wherein the patient interface comprises an acoustically-neutral elastic membrane,

wherein the acoustically-neutral elastic membrane is configured to inflate in order to couple to an alignment feature configured to receive a predetermined anatomical landmark in order to establish a location registration between at least two of the ultrasonic transducer array, the acoustically-neutral elastic membrane, and the anatomical structure,

wherein the cooling system comprises one or more cooling channels for receiving the fluid, wherein the one or more cooling channels are configured for circulation of the fluid to reduce heat at the acoustically-neutral elastic membrane; and

wherein the controller is coupled to the ultrasonic transducer array,

wherein the controller is configured to drive the ultrasonic transducer array with a signal configured to minimize a spatial variation of an acoustic wave intensity to cause the ultrasonic transducer array to activate a sonosensitizer in a treatment region in the anatomical structure with the planar or defocused acoustic waves at a frequency selected from a range of 20 kHz to 2 MHZ,

wherein the signal is configured with one or more duty cycles to drive each piezoelectric transducer in the ultrasonic transducer array to generate a high temporal peak acoustic intensity with a low temporal average acoustic intensity when activating the sonosensitizer at the treatment region in the anatomical structure with the planar or defocused acoustic waves.

11. The ultrasound transducer system of claim 10 , wherein the ultrasound transducer system comprises a user interface, and wherein the controller is configured to recommend a selected one of the plurality of treatment positions via the user interface.

12. The ultrasound transducer system of claim 10 , wherein the acoustically-neutral elastic membrane is a balloon or a gel layer, wherein the acoustically-neutral elastic membrane is positioned over an exit plane of the ultrasonic transducer array.

13. The ultrasound transducer system of claim 10 , wherein the anatomical structure is a zygomatic arch, a mastoid tip, or a middle arch of an eyebrow.

The ultrasound transducer system of claim 10 , wherein a volume of the fluid in the cavity is selectively configured to adjust in concert with at least one motion of the array holder to control a location of the ultrasonic transducer array and a distance of the ultrasonic transducer array with respect to the anatomical structure.
