Abstract

Disclosed herein are drug delivery devices and methods for the treatment of ocular disorders requiring targeted and controlled administration of a drug to an interior portion of the eye for reduction or prevention of symptoms of the disorder. The devices are capable of controlled release of one or more drugs and may also include structures which allow for treatment of increased intraocular pressure by permitting aqueous humor to flow out of the anterior chamber of the eye through the device.

Claims (17)

1. A drug delivery ocular implant comprising

an outer shell having a first end portion and a second end portion, said outer shell being shaped to define an interior space;

at least a first drug positioned within the interior space, wherein the at least the first drug is in the form of one or more pellets;

at least one partition positioned within the interior space to form a seal within the interior space;

one or more orifices positioned in the first end portion of the outer shell; and

a permeable material located wholly within the interior space in the first end portion of the outer shell and located between the at least the first drug and the one or more orifices; and

wherein the permeable material is permeable to the at least the first drug, thereby allowing passage of the at least the first drug through the permeable material, through the one or more orifices and out of the implant.

2. The implant of claim 1 , wherein said pellets have a minor axis of 0.28 to 0.31 mm and a major axis of 0.8 to 1.1 mm.

The implant of claim 1 , wherein the at least the first drug is present in an amount of at least 70% by weight of a total weight of the one or more pellets, wherein said pellets have a surface area to volume ratio of 13 to 17, and wherein said pellets have an aspect ratio of length to diameter of 2.8 to 3.6.

4. The implant of claim 1 , wherein said pellets are configured to balance osmotic pressure between said interior space and an ocular environment external to the implant after implantation.

5. The implant of claim 1 , wherein said pellets are coated with a polymeric coating that regulates the release of said first drug from said pellets.

The implant of claim 1 , wherein the at least the first drug is selected from the group consisting of prostaglandin, bimatoprost, latanoprost, travoprost, unoprostone, angiostatin, anecortave acetate, thrombospondin, VEGF receptor tyrosine kinase inhibitors and anti-vascular endothelial growth factor (anti-VEGF) drugs.

7. The implant of claim 6 , wherein said anti-VEGF drugs are selected from the group consisting of ranibizumab, bevacizumab, pegaptanib, sunitinib and sorafenib.

8. The implant of claim 1 , wherein said micro-tablets have a density of 0.7 g/cc to 1.6 g/cc.

9. The implant of claim 1 , wherein the at least one or more pellets comprises two or more pellets; wherein the at least one partition is positioned between the two or more pellets.

10. The implant of claim 9 , wherein the at least one partition is configured to bioerode.

11. The implant of claim 1 , wherein the at least one partition positioned within the forms a seal against the interior space to prevent drug elution in an unwanted direction.
