Abstract

Plants with reduced gluten grains and compositions thereof are disclosed herein.

Claims (21)

1. Wheat grain comprising: a wheat prolamin box binding factor (“WPBF”) gene with one or more non-transgenic human induced mutations encoding a WPBF protein comprising an amino acid sequence having 95% or greater identity to SEQ ID NO:3, or having 95% or greater identity to SEQ ID NO:6, or having 95% or greater identity to SEQ ID NO:9, wherein the grain comprises an increase in high molecular weight glutenin content as compared to grain from a wild type plant.

2. The wheat grain of claim 1 , wherein the grain further comprises a reduced gluten content as compared to grain from a wild type plant.

3. The wheat grain of claim 1 , wherein the grain further comprises a reduced gliadin content as compared to grain from a wild type plant.

4. The wheat grain of claim 1 , wherein the grain further comprises a reduced low molecular weight glutenin content as compared to grain from a wild type plant.

5. The wheat grain of claim 1 , wherein the grain further comprises an increase in lysine content as compared to grain from a wild type plant.

6. The wheat grain of claim 1 , wherein the WPBF protein comprises an amino acid sequence having 95% or greater identity to SEQ ID NO: 3.

7. The wheat grain of claim 1 , wherein the WPBF protein comprises an amino acid sequence having 95% or greater identity to SEQ ID NO: 6.

8. The wheat grain of claim 1 , wherein the WPBF protein comprises an amino acid sequence having 95% or greater identity to SEQ ID NO: 9.

9. Flour comprising a cell of the wheat grain of claim 1 .

10. A food or beverage product comprising a cell of the wheat grain of claim 1 .

11. A method of altering glutenin content in a wheat plant comprising;

(a) introducing at least one human induced mutation in a WPBF gene in plant material or plant parts from the wheat plant;

(b) growing the plant material or plant parts to produce progeny wheat plants; and

(c) selecting a progeny wheat plant that produces wheat grain having the one or more non-transgenic human induced mutations in the WPBF gene encoding a WPBF protein comprising an amino acid sequence having 95% or greater identity to SEQ ID NO: 3, or having 95% or greater identity to SEQ ID NO: 6, or having 95% or greater identity to SEQ ID NO: 9, wherein the grain comprises an increase in high molecular weight glutenin content as compared to grain from a wild type plant.

12. The method of claim 11 , wherein the grain further comprises a reduced gluten content as compared to grain from a wild type plant.

13. The method of claim 11 , wherein the grain further comprises a reduced gliadin content as compared to grain from a wild type plant.

14. The method of claim 11 , wherein the grain further comprises a reduced low molecular weight glutenin content as compared to grain from a wild type plant.

15. The method of claim 11 , wherein the grain further comprises an increase in lysine content as compared to grain from a wild type plant.

16. The method of claim 11 , wherein the WPBF gene comprises a nucleic acid sequence having 95% or greater identity to SEQ ID NO: 2.

17. The method of claim 11 , wherein the WPBF gene comprises a nucleic acid sequence having 95% or greater identity to SEQ ID NO: 5.

18. The method of claim 11 , wherein the WPBF gene comprises a nucleic acid sequence having 95% or greater identity to SEQ ID NO: 8.
