Abstract

In one aspect, a system for performing flow cytometry is disclosed, which comprises a laser for generating laser radiation for illuminating a sample, at least one detector for detecting at least a portion of a radiation emanating from the sample in response to said illumination so as to generate a temporal signal corresponding to said detected radiation, and an analysis module for receiving said temporal signal and performing a statistical analysis of said signal based on a forward model to reconstruct an image of said sample.

Claims (32)

1 . A system for performing flow cytometry, the system comprising:

a light source configured to concurrently illuminate a plurality of spatial locations on a particle as the particle passes through two or more discrete positions along a flow cell;

a detector configured to measure temporal waveform emissions that are generated from the illuminated particle at the two or more discrete positions along the flow cell;

a computer-readable storage medium comprising instructions that, when executed by one or more processing devices, cause the one or more processing devices to:

receive the temporal waveform emissions;

divide the temporal waveform emissions into a plurality of temporal segments;

generate image parameters of the particle based at least in part on the temporal segments of the temporal waveform emissions that are generated at the two or more discrete positions; and

display the image parameters of the particle.

2 . The system according to claim 1 , wherein the light source comprises a laser.

3 . The system according to claim 1 , wherein the light source comprises an LED.

4 . The system according to claim 1 , wherein the detector comprises a PMT.

5 . The system according to claim 1 , wherein the system further comprises a sorter for sorting the particle.

6 . The system according to claim 1 , wherein the instructions generate cell morphology data of the particle.

7 . The system according to claim 1 , wherein the image parameters comprise an image.

8 . The system according to claim 7 , wherein the image is any of a fluorescence, a darkfield, and a brightfield image.

9 . The system according to claim 1 , wherein the temporal waveform emissions are fluorescence signals.

10 . The system according to claim 1 , wherein the temporal waveform is a scattering signal.

11 . A flow cytometry method comprising:

concurrently illuminating with a light source a plurality of spatial locations on a particle as the particle passes through two or more discrete positions along a flow cell;

measure temporal waveform emissions that are generated from the illuminated particle with a detector at the two or more discrete positions along the flow cell;

divide the temporal waveform emissions into a plurality of temporal segments;

generate image parameters of the particle based at least in part on the temporal segments of the temporal waveform emissions that are generated at the two or more discrete positions; and

display the image parameters of the particle.

12 . The method according to claim 11 , wherein the light source comprises a laser.

13 . The method according to claim 11 , wherein the light source comprises an LED.

14 . The method according to claim 11 , wherein the detector comprises a PMT.

15 . The method according to claim 11 , wherein the method further comprises sorting the particle.

16 . The method according to claim 11 , wherein the method comprises generating cell morphology data of the particle.

17 . The method according to claim 11 , wherein the image parameters comprise an image.

18 . The method according to claim 17 , wherein the image is any of a fluorescence, a darkfield, and a brightfield image.

19 . The method according to claim 11 , wherein the temporal waveform emissions are fluorescence signals.

20 . The method according to claim 11 , wherein the temporal waveform is a scattering signal.
