Abstract

Among other things, two or more different antibodies are caused to bind to one or more units of a chemical component in a sample. Each of the antibodies is attached to one or more beads (e.g., microbeads). The sample is situated on a surface of an image sensor. At the image sensor, light is received originating at a light source that is other than the beads. The received light includes light reflected by, refracted by, or transmitted through the beads. At least one image of the sample is processed to separately enumerate individual beads and complexes of two or more of the beads attached to the two or more antibodies that are bound to a unit of the chemical component. The results of the processing are used to identify a presence or a level of the chemical component in the sample.

Claims (18)

1 . A computer-implemented method for identifying a presence or a level of a chemical component in a sample, the method comprising:

obtaining one or more images of the sample, the sample comprising (i) a plurality of multi-bead complexes, wherein each multi-bead complex comprises a unit of the chemical component bound to two or more beads, and (ii) a plurality of singleton beads that are not in multi-bead bound complexes with units of the chemical component;

determining a count of separate multi-bead complexes in the one or more images, wherein determining the count of separate multi-bead complexes comprises associating the two or more beads of each multi-bead complex of the plurality of multi-bead complexes with one another based on a proximity to one another of the two or more beads;

determining a count of singleton beads in the one or more images;

accessing a stored relationship between the count of separate multi-bead complexes, the count of singleton beads, and a concentration of the chemical component; and

identifying, based on the stored relationship, on the count of separate multi-bead complexes in the image, and on the count of singleton beads in the image, the presence or the level of the chemical component in the sample.

2 . The computer-implemented method of claim 1 , wherein, within each multi-bead complex, the two or more beads of the multi-bead complex are bound to the unit of the chemical component of the multi-bead complex by antibodies.

3 . The computer-implemented method of claim 1 , wherein the stored relationship comprises a standard curve.

4 . The computer-implemented method of claim 1 , wherein the stored relationship characterizes a test sample under incubation conditions matching incubation conditions of the sample.

The computer-implemented method of claim 1 , in which at least two beads of the two or more beads of a first multi-bead complex of the plurality of multi-bead complexes differ in at least one of appearance, reflective characteristics, refractive characteristics, or transmissive characteristics.

The computer-implemented method of claim 5 , in which the appearance, reflective characteristics, refractive characteristics, or transmissive characteristics comprise at least one of colors of the at least two beads, sizes of the at least two beads, shapes of the at least two beads, or birefringence of the at least two beads.

7 . The computer-implemented method of claim 1 , comprising causing a light source to emit light at the sample.

8 . The computer-implemented method of claim 7 , wherein the one or more images are based on the light

emitted by the light source,

reflected by, refracted by, or transmitted through the two or more beads of each multi-bead complex, and

received at an image sensor.

9 . The computer-implemented method of claim 1 , comprising causing movement of a first surface to confine the sample between the first surface and a second surface, wherein the sample is disposed on the second surface.

10 . The computer-implemented method of claim 9 , comprising causing the first surface to form a monolayer of the sample between the first surface and the second surface.
